Inhibition of small G proteins of the Rho family by statins orClostridium difficiletoxin B enhances cytokine-mediated induction of NO synthase II

2000

In order to investigate the involvement of Ras and/or Rho proteins in the induction of the inducible isoform of nitric oxide synthase (NOS II) we used HMG-CoA reductase inhibitors (statins) and Clostridium difficile toxin B (TcdB) as pharmacological tools. Statins indirectly inhibit small G proteins by preventing their essential farnesylation (Ras) and/or geranylgeranylation (Rho). In contrast, TcdB is a glucosyltransferase and inactivates Rho-proteins directly. Human A549/8- and DLD-1 cells as well as murine 3T3 fibroblasts were preincubated for 18 h with statins (1–100 μM) or TcdB (0.01–10 ng ml−1). Then NOS II expression was induced by cytokines. NOS II mRNA was measured after 4–8 h by R…

Lovastatin inhibits Rho-regulated expression of E-selectin by TNFalpha and attenuates tumor cell adhesion.

2003

E-selectin mediated cell-cell adhesion plays an important role in inflammatory processes and extravasation of tumor cells. Tumor necrosis factor-alpha (TNF-alpha) induces E-selectin gene and protein expression in primary human endothelial cells (HUVEC) and in an endothelial cell line (EA.hy-926). As shown by ELISA and FACS analyses, HMG-CoA reductase inhibitors (e.g., lovastatin) impair the TNF-alpha stimulated increase in E-selectin protein expression. Similar results were obtained for E-selectin mRNA expression and promoter activity, indicating that the effect of lovastatin is based on inhibition of gene expression. The effective inhibitory concentration of lovastatin was in a physiologic…

Online expression and spending on personal cybersecurity

2019

The Internet is used increasingly as a platform both for free expression and e-commerce. Internet users have a variety of attitudes towards the security and privacy risks involved with using the Internet; and distinct concerns and behaviors with regard to expressing themselves online. Users may have controversial viewpoints that they may express online in various ways. Controversial viewpoints or artwork by their nature may not be as well received as positive or polite expressions. In the online environment, users with controversial viewpoints may be reluctant to express the viewpoints due to concern about possible consequences resulting from the expressions. Consequences may be imposed by …

L’enseignement des expressions figées par le biais du roman graphique Persépolis

2019

The constant search for suitable teaching materials to achieve the linguistic skills provided by a language teaching course, finds in authentic documents an important instrument for teaching foreign languages. The scientific literature attesting to the effectiveness of the authentic material is wide and consolidated; indeed, a text conceived by a native for a native, and, therefore, not developed for the sole purpose of teaching a foreign language, is intrinsically carrying a cultural richness and a linguistic variety, which are rarely present in any didactic artifact. The authentic material therefore represents an opportunity to meet the fixed expressions that are part of the communicative…

The Paracentrotus lividus metallothionein gene family: structure and expression.

2014

Metallothioneins are metal binding proteins that play a pivotal role in metal homeostasis and detoxification. Since their initial discovery, they have been extensively studied in a variety of organisms ranging from microbes to plants and animals. Organisms often possess multiple genes encoding metallothionein homologs with distinct properties, such as varying affinities for different metals, and in many cases different functions. Despite the plethora of available studies, very little information is known about sea urchin P. lividus MT (1, 2). We previously identified five Pl-MT embryonic cDNAs and we studied their induction after cadmium treatment (3). Now we studied their expression during…

Chromatin dynamics during sea urchin embryogenesis: effects on the neural alpha tubulin PlTa2 gene expression

2011

Expression of PlTa2 gene during sea urchin P. lividus development, is spatially confined to the neural territory and temporally activated from the blastula stage. To evaluate a possible involvement of chromatin modifications in regulation of PlTa2 gene expression we first searched for DNaseI hypersentive sites. We found four sites localized in the introns of the gene, when we used chromatin extracted from embryos at gastrula stage but not from morula stage. This result suggests a possible functional role of the introns in the activation of the expression of PlTa2 gene. Moreover, we used specific antibodies for RNA polymerase II and for different modified form of lysine 9, lysine 27 and lysi…

Bcl-xL as a Modulator of Senescence and Aging

2021

Many features of aging result from the incapacity of cells to adapt to stress conditions. When cells are overwhelmed by stress, they can undergo senescence to avoid unrestricted growth of damaged cells. Recent findings have proven that cellular senescence is more than that. A specific grade of senescence promotes embryo development, tissue remodeling and wound healing. However, constant stresses and a weakening immune system can lead to senescence chronicity with aging. The accumulation of senescent cells is directly related to tissue dysfunction and age-related pathologies. Centenarians, the most aged individuals, should accumulate senescent cells and suffer from their deleterious effects,…

Multi-word Lexical Units Recognition in WordNet

2022

WordNet is a state-of-the-art lexical resource used in many tasks in Natural Language Processing, also in multi-word expression (MWE) recognition. However, not all MWEs recorded in WordNet could be indisputably called lexicalised. Some of them are semantically compositional and show no signs of idiosyncrasy. This state of affairs affects all evaluation measures that use the list of all WordNet MWEs as a gold standard. We propose a method of distinguishing between lexicalised and non-lexicalised word combinations in WordNet, taking into account lexicality features, such as semantic compositionality, MWE length and translational criterion. Both a rule-based approach and a ridge logistic regre…

Effects of ectopic expression of NGAL on doxorubicin sensitivity.

2012

Neutrophil gelatinase-associated lipocalin (NGAL, a.k.a Lnc2) is a member of the lipocalin family which has diverse roles including stabilizing matrix metalloproteinase-9 from auto-degradation and as siderocalins which are important in the transport of iron. NGAL also has important biological functions involved in immunity and inflammation as well as responses to kidney damage. NGAL expression has also been associated with certain neoplasia and is important in the metastasis of breast cancer. Many advanced cancer patients have elevated levels of NGAL in their urine and it has been proposed that NGAL may be a prognostic indicator for certain cancers (e.g. breast, brain, and others). NGAL exp…
