Bacteria consortia and deterioration of archaeological waterlogged wood: identification by molecular and microscopy techniques

In this study molecular tools are applied to reveal and identify bacterial colonization in waterlogged wood to assessing the changes induced in anatomical structure, previously observed by Optical and Scanning Electron Microscopy (1). The results obtained by observation of wooden thin sections (OM), shown the presence of black and dark-brown areas and mineral concretions. The SEM analysis revealed a specific cell walls alteration, attributable to bacterial activity, other than abundant pyrite framboids (FeS2). Studies on methylation of PTHrP gene promoter 2 performed on five CpG island internal sites using the Methylation-Sensitive Restriction Endonuclease Multiplex (MSREM)-PCR showed that one of the sites nearest the trascription starting point is heavy methylated in a significantly high number of RA patients. Thus RA seems to be characterized by genomewide hypomethylation associated with local hypermethylation like the most part of tumors. This result raises the possibility that susceptibility to lymphomas is related to abnormal DNA methylation levels and suggests the opportunity to evaluate the DNA methylation status in RA patientin fact the demethylating therapies together with diet and life style can act towards an increase of tumor risk. Future studies using a larger number of subjects could confirm these findings.Rheumatoid Arthritis (RA) is a chronic multisystem inflammatory disease characterized by high recurrence of lymphomas as well as hypercalcemia due to PTHrP overexpression. Because of DNA methylation plays a critical role in development of neoplasias we determined in RA patients the global DNA methylation status and local methylation pattern of the CpG island of one of the three promoters of PTHrP gene utilizing molecular and cytogenetic techniques. Investigations performed on DNA from peripheral blood of patients and donors amplified by Methylation-Sensitive Arbitrarily Primed (MeS-AP)-PCR indicated that RA is strongly associated with global DNA hypomethylation. This site, also known as Monasterium Album, has a long and tormented history linked with volcanic activity of Mt. Etna and regional seismicity. Microstructure and mechanical behaviour of a saturated compacted scaly clay

The paper presents the results of a research on the influence of structure characteristics on the mechanical behaviour of a saturated compacted scaly clay. To this aim, mechanical properties of compacted scaly clay samples after saturation are compared with the corresponding properties measured on the clay reconstituted in the laboratory. For the stress range explored in the experimental programme, compacted scaly clay samples are stiffer and stronger than reconstituted clay samples owing to their greater density. The welding of the inclined web bars to top bars, both of steel type B450C, and to bottom steel plate, the latter made of S355 steel type, are tested. Experimental investigation on high-strength fibre-reinforced concrete beams subjected to bending and shear

The results of an experimental campaign carried out on a number of high-strength concrete beams loaded in 4-point bending are presented in this paper, where the roles of the both steel hooked fibres and the transverse reinforcement are investigated, for different values of the shear span. All tests were displacement-controlled. The results – mostly in terms of load-deflection curves – confirm what is well known in the literature, that an adequate amount of steel fibres can turn a brittle shear-type failure into a ductile flexural-type failure, to the advantage of the ultimate bearing capacity in flexure and shear, that can be fully exploited without increasing the transverse reinforcement. …
