New Lipid Modulating Drugs: The Role of Microsomal Transport Protein Inhibitors

Microsomal triglyceride transfer protein (MTP) is involved in the synthesis of very low density lipoprotein in the liver. Its deficiency results in abetalipoproteinemia. MTP inhibitors target the assembly and secretion of apolipoprotein B-containing lipoproteins. These agents may potentially play a role, alone or in combination, in the treatment of hypercholesterolemia or hypertriglyceridemia. Clinical applications of MTP inhibitors initially focused primarily on high-dose monotherapy in order to produce substantial reductions in LDL-cholesterol levels but these proved to induce significant hepatic steatosis and transaminase elevations. Combined treatment with trypsin, cholesterol esterase, and neuraminidase transforms LDL, but not HDL or VLDL, to particles with properties akin to those of lipid extracted from atherosclerotic lesions. Single or double enzyme modifications, or treatment with phospholipase C, or simple vortexing are ineffective. Improvement of photo-stability of LLDPE-based nanocomposites

LLDPE-based nanocomposites undergo faster photo-oxidation than the unfilled matrix and the extent of their physical properties deterioration, namely elongation at break and tensile strength, is dependent on the organo-nanoclay loading. The observed acceleration is not due to faster photo-oxidation but rather due to a reduction of the oxidation induction time. The presence in the organoclays of trace amounts of metal ions may be promoting catalytic photodegradation.
