Immunoelectron microscopy labeling for the identification of oligodendrocytes and their precursor cells

2021

A new efficient, highly dispersed, Pd nanoparticulate silica supported catalyst synthesized from an organometallic precursor. Study of the homogeneou…

2018

[EN] A new Pd(0) catalyst supported on silica UVM-7 has been synthesized from the organometallic [Pd-2(mu-(C6H4) Pp(2))(2)(CH3CN)(4)](BF4)(2) precursor, characterized by the high dispersion, activity, and small size of the palladium nanoclusters fixed on the silica surface. The catalyst was tested for the Suzuki-Miyaura (SM) reaction of different 4-substitutedphenyl halides with phenylboronic acid. The kinetic study concurs with most of the catalytic action was carried out by Pd species originated by the partial solubilization of Pd immobilized on mesoporous silica. The Schmidt's analysis of differential selectivity (SADS) in several competitive SM reactions, together the STEM-HAADF and HRT…

Classification par méthodes d’apprentissage supervisé et faiblement superviséd’images multimodales pour l’aide au diagnostic du lentigo malin en derm…

2021

Carried out in collaboration with the Saint-Étienne University Hospital, this work provides additional information to help the skin diagnosis by providing new decision methods on Lentigo Maligna and Lentigo Maligna Melanoma pathologies. To this end, the modalities regularly used in clinical conditions are made available to this work and are orchestrated within a multimodal process. Among image modalities, may be mentioned the clinical photography, the dermatoscopy, and the confocal reflectance microscopy. Initially, the first steps of this manuscript focus on reflectance confocal microscopy as the work in computer diagnostic assistance is relatively underdeveloped, in particular on the dete…

Molecular imaging of VEGF in gastrointestinal cancer in vivo using confocal laser endomicroscopy

2010

Vascular endothelial growth factor (VEGF) is a therapeutic target in gastrointestinal cancer (GiC). However, its in vivo visualisation could not be achieved to date with endoscopic techniques. Confocal laser endomicroscopy (CLE) is a novel imaging technique for gastrointestinal endoscopy providing in vivo microscopy at subcellular resolution. The aim of the study was to evaluate CLE for in vivo molecular imaging of VEGF in GiC.Molecular imaging of tumours in APCmin mice, in xenograft models and in surgical specimens of patients with colorectal cancer (CRC) was achieved after application of labelled antibodies. The tumour sites were scanned with the probe for the strongest specific fluoresce…

Agonist and antagonist-dependent internalization of the human vasopressin V2 receptor.

1998

Abstract In this report we demonstrate that in HEK293 cells stably expressing the human V2vasopressin receptor, ligand-induced internalization of the hormone receptor occurs via the clathrin-dependent pathway. Studies of receptor trafficking either by direct visualization of the V2receptor by confocal microscopy or binding experiments show a rapid internalization (half-time 6–7 min). Blocking of the clathrin-dependent pathway by hypertonic sucrose increased vasopressin-induced cellular cAMP production and decreased the desensitization of the V2receptor–adenylyl cyclase system. Thus, internalization appears to be a major regulatory mechanism terminating vasopressin action in HEK293 cells. Tw…

Immunoelectron Microscopy of Vesicle Transport to the Primary Cilium of Photoreceptor Cells

2009

Cilia are organelles of high structural complexity. Since the biosynthetic machinery is absent from cilia all their molecular components must be synthesized in organelles of the cytoplasm and subsequently transported to the cilium. Ciliary cargos are thought to be translocated in the membrane of transport vesicles or association with these vesicles to the base of the cilium where the vesicles fuse with the periciliary target membrane for further delivery of their cargo into the ciliary compartment by the intraflagellar transport (IFT). Here we describe a modified preembedding labeling method as an alternative technique to conventional postembedding methods eligible for analyses of ciliary c…

Fast Axial Scanning in 3D Imaging

2016

We establish the conditions for obtaining fast axial 3D microscopy with constant magnification, invariant resolution, and symmetrical displacement range. We demonstrate that liquid lenses permit the axial scanning free of mechanical movements and vibrations. This allows speeding-up the process and avoiding distortionsin the image of dynamic specimens. Activity investigation of pinostrobin towards herpes simplex virus-1 as determined by atomic force microscopy

2009

In the present study, the antiviral activity of pinostrobin towards herpes simplex virus-1 (HSV-1) was investigated by MTT assay and atomic force microscopy. Pinostrobin can inhibit HSV-1 replication with 50% effective concentration (EC(50)) of 22.71 ± 1.72 μg/ml. MTT assay showed HSV-1 was significantly inhibited when pretreated with pinostrobin, with the inhibition of 85.69 ± 2.59%. Significant changes in morphology and size of HSV-1 were observed by atomic force microscopy (AFM) in response to pinostrobin treatment. AFM topography and phase images showed that with increasing time, the envelope was shedded and damaged, finally leading to virus inactivation. With increasing concentration, …

How and why does willow biochar increase a clay soil water retention capacity?

2018

Addition of biochar into a soil changes its water retention properties by modifying soil textural and structural properties. In addition, internal micrometer-scale porosity that is able to directly store readily plant available water affects soil water retention properties. This study shows how precise knowledge of the internal micrometer-scale pore size distribution of biochar can deepen the understanding of the biochar-water interactions in soils. The micrometer-scale porosity of willow biochar was quantitatively and qualitatively characterized using X-ray tomography, 3D image analysis and Helium ion microscopy. The effect of biochar application on clay soil water retention was studied by…
