Thermodynamics and kinetics of ion translocation in the human wild-type and E-1_A α7 nicotinic receptor

We use an all-atom model of the human nicotinic acetylcholine receptor α7 in a conductive conformation, to provide the first available mapping of the potential of mean force for the ion translocation across the channel. The modeling is based on MD simulations combined with the milestoning method with Voronoi tessellation. The quality of the protein model and description is confirmed by the agreement with experimental data for proteins of the same family. The specific mutation E-1 ' A at the cytoplasmatic filter is here shown to strongly affect both sodium and chloride permeation, leading to a complete inversion of selectivity.

Abstract Introduction Red yeast rice (RYR) is an effective cholesterol-lowering nutraceutical reversibly inhibiting 3-hydroxy-3-methyl-glutaryl-CoA (HMG-CoA) reductase. As liver damage is a possible (albeit rare) side effect of HMG-CoA inhibitors, it make sense to focus on the tolerability of the liver to RYR extracts. The aim of this paper is to offer an expert opinion on the risk of liver damage by the use of RYR extract. The septal organ (SO) is a small island of olfactory epithelium located at the ventral base of the nasal septum. SO neurons express identified odorant receptors, but its function remains unknown. Using perforated patch clamp recordings, we investigated the response properties of SO neurons in the intact epithelium of mice to a panel of 45 odorants or mixtures. Out of 328 neurons tested, approximately 70% responded to odor stimulation. Among the responsive neurons, 72.5 % responded to multiple odorants. Responding neurons were tested with increasing concentrations of 3 odorants (octanoic acid, (+) camphor, amylacetate) and a mixture (fox urine). Dose-response curves show nanomolar threshold …
