Sperm DNA integrity and human papillomavirus (HPV) infections: a controversy that could be resolved by a new molecular approach

Study question: The aim was to determine if HPVs affect spermatozoa DNA integrity. To resolve the discrepancy regarding the association between HPV infections and sperm DNA damage. Summary answer: To elucidate if HPV impairs DNA integrity, we suggest to investigate HPV DNA positivity in spermatozoa by a differential lysis procedure before TUNEL assay.

Gene expression in mouse spermatogenesis during ontogenesis

In this study, we evaluated the expression of genes probably involved in spermatogenesis in the mouse. We examined cytosolic chaperonin theta subunit (CCTtheta), Ngg1 interacting factor 3 like 1 binding protein 1 (NIF3L1 BP1) and apolipoprotein H (ApoH) expression during mouse onto-geny using RT-PCR. Testicular tissue was obtained from mice 3, 6, 8, 10, 12, 14, 18, 20 and 40 (adult) days after birth. For each mouse, one testis was used for histological examination, whereas RNA was extracted from the controlateral testis for expression analysis. New molecular markers for the evaluation of gamete quality.

Purpose: Only 30 % of IVF cycles result in a pregnancy, so that multiple embryos need to be replaced, per treatment cycle, to increase pregnancy rates, resulting in a multiple gestation rate of 25 %. The use of new markers in the gamete selection, could reduce the number of the oocytes to be fertilized and embryos to be produced, but the tools to evidence the gamete competence remain unavailable and more studies are needed to identify bio-markers to select the best oocyte and sperm to produce embryos with higher implantation potentiality.
