Toll-like receptor 2 is dispensable for acquired host immune resistance to Candida albicans in a murine model of disseminated candidiasis

2004

Previous work by our group showed that Toll-like receptor 2 (TLR2) is essential for activation of innate immunity, playing a major role in the response of macrophages to Candida albicans, triggering cytokine and chemokine expression, and therefore TLR2 -/- mice are more susceptible to systemic primary candidiasis. In this work, we used a murine model of systemic C. albicans infection, in which resistance to reinfection with virulent wild-type cells is induced by prior exposure of mice to a low-virulence agerminative strain of C. albicans (primary sublethal infection), to study the influence of TLR2 gene deletion on (i) the ability to develop an acquired resistance upon vaccination; (ii) the…

Toll-like receptor-2 is essential in murine defenses against Candida albicans infections

2004

In this work, we studied the role of toll-like receptor-2 (TLR2) in murine defenses against Candida albicans. TLR2-deficient mice experimentally infected intraperitoneally (i.p.) or intravenously (i.v.) in vivo had very significant impaired survival compared with that of control mice. In vitro production of TNF-alpha and macrophage inhibitory protein-2 (MIP-2) by macrophages from TLR2-/- mice in response to yeasts and hyphae of C. albicans were significantly lower (80% and 40%, respectively; P <0.05) than production by macrophages from wild-type mice. This impaired production of TNF-alpha and MIP-2 probably contributed to the 41% decreased recruitment of neutrophils to the peritoneal cavity…

TLR3-induced activation of mast cells modulates CD8+ T-cell recruitment.

2005

AbstractMast cells play an important role in host defense against various pathogens, but their role in viral infection has not been clarified in detail. dsRNA, synthesized by various types of viruses and mimicked by polyinosinic-polycytidylic acid (poly(I:C)) is recognized by Toll-like receptor 3 (TLR3). In this study, we demonstrate that poly(I:C) injection in vivo potently stimulates peritoneal mast cells to up-regulate a number of different costimulatory molecules. Therefore, we examined the expression and the functional significance of TLR3 activation in mast cells. Mast cells express TLR3 on the cell surface and intracellularly. After stimulation of mast cells with poly(I:C) and Newcas…

Quantitative determination of surface area of silica gel particles by near infrared spectroscopy and chemometrics

2008

Abstract Surface areas of silica gel particles have been quantitatively determined by using near infrared spectrometry and chemometrics. Surface areas of six silica gel samples with varying surface area (300–750 m2/g) have been determined by the common Brunauer–Emmett–Teller (BET) method and used in preparing mixtures of silica gel samples with varying surface area in the ranges 300–474 and 500–750 m2/g. The near infrared spectra of the mixtures were measured and the spectral profiles calibrated against the calculated surface area of the mixtures as dependent variable using partial least squares (PLS) technique. The models obtained from the calibrations were then used in predicting the surf…

Negative pressure cavitation-microwave assisted preparation of extract of Pyrola incarnata Fisch. rich in hyperin, 2'-O-galloylhyperin and chimaphili…

2013

Abstract A novel and effective extraction method, namely negative pressure cavitation-microwave assisted extraction technique (NMAE), was developed for the preparation of extracts of Pyrola incarnata Fisch., which are rich in the main constituents hyperin, 2′-O-galloylhyperin and chimaphilin. Single factor experiments and Box–Behnken design (BBD) were combined with a response surface methodology to examine factors affecting extraction. Maximum extraction yields of hyperin, 2′-O-galloylhyperin and chimaphilin (1.339 ± 0.029, 4.831 ± 0.117 and 0.329 ± 0.011 mg/g, respectively) were achieved under the following optimised conditions: 700 W microwave power, 50 °C extraction temperature, 30:1 mL/…

Supramolecular PEG-co-oligo(p-benzamide)s prepared on a peptide synthesizer.

2007

An automated synthesis protocol has been developed for the preparation of oligo(p-benzamide)s on solid support using a commercial peptide synthesizer employing a variation of standard Fmoc chemistry. Bis(trichloromethyl carbonate) in NMP was used to activate the aromatic carboxylic acids for acylation of secondary aromatic amines on solid support. N-Protected hepta(p-benzamide) was automatically prepared on solid support and manually converted to a solid supported block co-oligomer by attaching a poly(ethylene glycol) chain. Cleavage from the support could be achieved with minimal loss of the p-methoxybenzyl N-protective group. While the N-protected block co-oligomer was molecularly dissolv…

Comparison of MODIS and Landsat-8 retrievals of Chlorophyll-a and water temperature over Lake Titicaca

2016

Chlorophyll-a concentration ([Chl-a]) and Lake Surface Temperature (LST) were retrieved in Lake Titicaca (Peru-Bolivia) using MODIS and Landsat-8 images. The lake was chosen as a case-study for evaluating the feasibility of Landsat-8 images for [Chl-a] and LST monitoring in oligotrophic and mesotrophic water bodies. The big size of the lake and its spatial and temporal variability, allowed the comparison of MODIS and Landsat-8 products for a wide range of [Chl-a] and LST. The atmospheric correction of the images was facilitated by the very high altitude of the lake. MODIS images were processed with standard ocean color algorithms whereas for Landsat-8, specific algorithms were tested and va…

Optically Detected Magnetic Resonance of Chlorophyll Triplet States in Water-Soluble Chlorophyll Proteins from Lepidium virginicum: Evidence for Exci…

2018

Optically detected magnetic resonance of triplet states populated by photoexcitation in water-soluble chlorophyll proteins (WSCPs) from Lepidium virginicum has been performed using both absorption and fluorescence detection. Well resolved triplet-singlet (T-S) spectra have been obtained and interpreted in terms of electronic interactions among the four chlorophylls (Chls), forming two dimers in the WSCP tetramer. Localization of the triplet state on a single Chl leads to a redistribution of the oscillator strength in the remaining three Chls of the complex. By comparing the spectra with those obtained on a substoichiometric WSCP complex containing only 2 Chls per protein tetramer, we proved…

Preliminary Analysis on Correlations between Spatial Distribution of Chlorophyll-a and Experimental Data of Biomass in the Strait of Sicily

2010

This study, using both remotely sensed and measured in situ data, is directed to the analysis of the correlations between the chlorophyll-a concentration and the biomass of sardines and anchovies acoustically evaluated in the Strait of Sicily. This work, inter alia, shows the usefulness of remote observation of seas in determining possible relationships between fish stocks and some oceanographic parameters (Sea Surface Temperature, Chlorophyll-a, Zooplankton).

Negative pressure cavitation accelerated processing for extraction of main bioactive flavonoids from Radix Scutellariae

2011

Abstract To enhance the extraction efficiency and reduce the energy consumption, an emerging technology named negative pressure cavitation extraction (NPCE) has been shown to be a feasible option for the extraction of bioactive compounds in agricultural crops and medicinal plants. Meanwhile, it can be applied at the pilot scale as a manufacturing process for edible and medicinal plants. Currently, NPCE was proposed for extraction of baicalin, wogonoside, baicalein and wogonin from Radix Scutellariae on the basis of a central composite design (CCD) and response surface methodology (RSM). With proper optimization (80 mesh of particle size, 40 mL/g of liquid/solid ratio, 75% aqueous ethanol as…
