Complexes of tetra(parasulphonatephenyl)porphyrinato ferrate(III), FeTPPs, with 1,10 phenanthroline and 2,2′ bipyridine ligands

Abstract Iron(III) porphyrin, FeTPPS, gives high-spin 1:1 monomeric complexes with the ligands 1,10 phenanthroline, 2,9 dimethylphenanthroline and 2,2 bipyridine in aqueous solution. Absorption spectrum of the complexes is very different to the one of the monomer metalloporphyrin, but nearly identical to the visible spectrum of the μ-oxo dimer Fe2(TPPS)2O. A spectrophotometric study of the formation equilibria yields the following values of the stability constants: Me2phen 4.3 × 104, phen 1.1 × 104, bipy 2.0 × 102 (25 °C, 0.1 M NaNO3). We have previously shown that hypoxia and N-methyl-D-aspartate (NMDA) receptor activation induce breakdown of choline-containing phospholipids in rat hippocampus, a process which is mediated by calcium influx and phospholipase A (2) activation. Bilobalide, a constituent of Ginkgo biloba, inhibited this process in a potent manner (Weichel et al., Naunyn-Schmiedeberg's Arch. Pharmacol. 360, 609-615, 1999). In this study, we used fluorescence microscopy and radioactive flux measurements to show that bilobalide does not interfere with NMDA-induced calcium influx. We have used two tumor cell clones (B9 and G2), derived from the methylcholanthrene-induced murine fibrosarcoma GR9 and normal BALB/c3T3 fibroblasts, to study the ability of t-BOOH derived reactive oxygen radicals to induce oxidative stress, apoptosis and c-fos and c-jun mRNA transcription. These clones differ in terms of their major histocompatibility complex (MHC) (H-2) class I genes expression, their tumor induction and metastatic potential and their reduced glutathione (GSH) levels. The TAR sequence of the 5' leader of HIV-1 long terminal repeat-directed mRNA was found to be able to bind to and to activate double-stranded RNA-dependent (2'-5')A synthetase. Binding of TAR to the purified synthetase in vitro was abolished by addition of HIV-1 Tat protein, which binds to this sequence with a high affinity. Inhibition of TAR-mediated activation of (2'-5')A synthetase by Tat was prevented in the presence of the Zn2+ and Cd2+ chelators o-phenanthroline and penicillamine, which did not impair TAR-synthetase interaction. Abstract Single crystals of the compound of formula [Ni(bipy)3]Cl2·5.5H2O (bipy=2,2′-bipyridine) have been prepared and structurally characterised by X-ray diffraction. The structure analysis reveals that the complex has an approximate D3 symmetry. The coordination sphere consists of six nitrogen atoms of the three-bipyridine ligands in an octahedral arrangement about the nickel. One of the chlorine ions is surrounded by twelve CH⋯Cl hydrogen bonds. Three kinds of layers are found in the crystal structure: (i) the Λ-[Ni(bipy)3]2+ cation; (ii) the water/chloride positions (O(1w)/Cl(21) to O(7w)/Cl(27)); (iii) the Δ-[Ni(bipy)3]2+ cation and a chloride ion (Cl(1)). Deprotonation of the ionogenic tetradentate ligand 6,6′-bis(1H-tetrazol-5-yl)-2,2′-bipyridine [H2bipy(ttr)2] in the presence of FeII in solution has afforded an anionic mononuclear complex and a neutral two-dimensional coordination polymer formulated as, respectively, NEt3H{Fe[bipy(ttr)2][Hbipy(ttr)2]}·3MeOH (1) and {Fe[bipy(ttr)2]}n (2). The anions [Hbipy(ttr)2]− and [bipy(ttr)2]2– embrace the FeII centers defining discrete molecular units 1 with the FeII ion lying in a distorted bisdisphenoid dodecahedron, a rare example of octacoordination in the coordination environment of this cation.
