321. Sea Urchin sns Chromatin Insulator Prevents Silencing and Positional Effect Variegation of Oncoretroviral Vectors Transgene Expression in Murine…
2006
Silencing and position effect are considered significant obstacles to obtain a consistent level of transgene expression in viral gene therapy. Furthermore recent studies had shown that retroviruses tend to land on active genes with the potential consequence of insertional mutagenesis. The inclusion of elements, such as chromatin insulators, capable to insulate a gene from the surrounding chromatin effects at the integration site should improve both efficacy and safety of gene therapy vectors. We have previously characterized a 265 bp insulator element, termed sns, localized at the 3’ end of the early histone H2A gene of the sea urchin Paracentrotus lividus. This sequence contains three cis-…
Mangifera indica L. Extract and Mangiferin Modulate Cytochrome P450 and UDP-Glucuronosyltransferase Enzymes in Primary Cultures of Human Hepatocytes
2012
The aqueous stem bark extract of Mangifera indica L. (MSBE) has been reported to have antioxidant, anti-inflammatory and analgesic properties. In previous studies, we showed that MSBE and mangiferin, its main component, lower the activity of some cytochrome P-450 (P450) enzymes in rat hepatocytes and human liver microsomes. In the present study, the effects of MSBE and mangiferin on several P450 enzymes and UDP-glucuronosyltransferases (UGTs) in human-cultured hepatocytes have been examined. After hepatocytes underwent a 48-h treatment with sub-cytotoxic concentrations of the products (50-250 µg/mL), a concentration-dependent decrease of the activity of the five P450 enzymes measured (CYP1A…

Prevention by Dietary Polyphenols (Resveratrol, Quercetin, Apigenin) Against 7-Ketocholesterol-Induced Oxiapoptophagy in Neuronal N2a Cells: Potentia…
2020
The Mediterranean diet is associated with health benefits due to bioactive compounds such as polyphenols. The biological activities of three polyphenols (quercetin (QCT), resveratrol (RSV), apigenin (API)) were evaluated in mouse neuronal N2a cells in the presence of 7-ketocholesterol (7KC), a major cholesterol oxidation product increased in patients with age-related diseases, including neurodegenerative disorders. In N2a cells, 7KC (50 µ
RNA-binding ability of PIPP in requires the entire protein
2003
Post-transcriptional fate of eukaryotic mRNAs depends on association with different classes of RNA-binding proteins (RBPs). Among these proteins, the cold-shock domain (CSD)-containing proteins, also called Y-box proteins, play a key role in controlling the recruitment of mRNA to the translational machinery, in response to environmental cues, both in development and in differentiated cells. We recently cloned a rat cDNA encoding a new CSD-protein that we called PIPPin. This protein also contains two putative double-stranded RNA-binding motifs (PIP(1) and PIP(2)) flanking the central CSD, and is able to bind mRNAs encoding H1 degrees and H3.3 histone variants. In order to clarify the role of…
Robustness of the Gaussian concentration inequality and the Brunn–Minkowski inequality
2016
We provide a sharp quantitative version of the Gaussian concentration inequality: for every $r>0$, the difference between the measure of the $r$-enlargement of a given set and the $r$-enlargement of a half-space controls the square of the measure of the symmetric difference between the set and a suitable half-space. We also prove a similar estimate in the Euclidean setting for the enlargement with a general convex set. This is equivalent to the stability of the Brunn-Minkowski inequality for the Minkowski sum between a convex set and a generic one.
Analytic Bergman operators in the semiclassical limit
2018
Transposing the Berezin quantization into the setting of analytic microlocal analysis, we construct approximate semiclassical Bergman projections on weighted $L^2$ spaces with analytic weights, and show that their kernel functions admit an asymptotic expansion in the class of analytic symbols. As a corollary, we obtain new estimates for asymptotic expansions of the Bergman kernel on $\mathbb{C}^n$ and for high powers of ample holomorphic line bundles over compact complex manifolds.
Use of HFC fluids as suitable replacements in low-temperature refrigeration plants
2014
An experimental investigation of the performance of a low-temperature refrigerating unit working with R22 and a comparison of its performance when operating with replacement HFC fluids in accordance with the European Regulation CE-1005/2009 are presented in this paper. Plant working efficiency was tested with R22, as baseline, and then compared with four different HFC fluids: R413A, R417A, R422A and R422D. The refrigerating unit was a vapour-compression plant equipped with a reciprocating double-cylinder compressor able to keep the cold room at -20ºC. Lower values of the temperature at the end of compression and polytrophic exponent can be achieved with the HFC tested. Substituting the R22 …
(Table 1) Sea surface temperature reconstruction for eastern equatorial Pacific surface sediment samples
2012
Significant uncertainties persist in the reconstruction of past sea surface temperatures in the eastern equatorial Pacific, especially regarding the amplitude of the glacial cooling and the details of the post-glacial warming. Here we present the first regional calibration of alkenone unsaturation in surface sediments versus mean annual sea surface temperatures (maSST). Based on 81 new and 48 previously published data points, it is shown that open ocean samples conform to established global regressions of Uk'37 versus maSST and that there is no systematic bias from seasonality in the production or export of alkenones, or from surface ocean nutrient concentrations or salinity. The flattening…
