Altered expression of inflammation-related genes in human carotid atherosclerotic plaques.

2011

Abstract Objective Inflammation is a pivotal process in atherosclerosis development and progression, but the underlying molecular mechanisms remain largely obscure. We have conducted an extensive expression study of atherosclerotic plaques to identify the inflammatory pathways involved in atherosclerosis. Methods We studied 11 human carotid plaques, their respective adjacent regions and 7 control arteries from different subjects. Expression of 92 genes was studied by TaqMan low-density array human inflammation panel. Human aortic endothelial and smooth muscle cells were used for in vitro experiments. Results The mRNA levels of 44/92 genes (48%) differed significantly between the tissues exa…

In vitro pro-apoptotic and anti-migratory effects of Treculia africana Decne. (Moraceae) and Entandrophragma angolense Welw (Meliaceae) extracts on p…

2021

Abstract Prostate cancer is the second leading cause of cancer deaths in men and there is an increasing interest in chemoprevention to fight it. The authors sought a scientific rationale for the traditional use of six Cameroonian medicinal plants for the treatment of prostate inflammation/tumour. The prostate cells viability incubated with each ethanolic plant extract was determined after 24, 48 and 72 h using the MTT assay. The antitumor mechanisms of promising extracts [Treculia africana (TA) and Entandrophragma angolense (EA)] were further assessed by evaluating cell growth, cell proliferation, cell cycle, cell death mechanisms and cell migration. Only TA and EA significantly inhibited L…

Role of cAMP in mediating AHR signaling.

2009

Regulation of the nuclear import of many transcription factors represents a step in gene regulation which is crucial for a number of cellular processes. The aryl hydrocarbon receptor (AHR), a basic helix-loop-helix protein of the PAS (PER-ARNT-SIM) family of transcriptional regulators is a cytosol-associated and ligand-activated receptor. The environmental toxin dioxin binds with high affinity to AHR rendering it nuclear and leading to the activation of AHR sensitive genes. However, the fact, that the AHR mediates a large variety of physiological events without the involvement of any known exogenous ligand, including liver and vascular system development, maturation of the immune system, re…

Neurosteroidogenesis in Rat Retinas

2002

Neurosteroids (steroids synthesized in the CNS) function by modulating neurotransmission. To establish an experimental model for investigation of neurosteroid synthesis and regulation, independent of blood-borne steroids, we examined the steroidogenic activity of isolated rat retinas. We identified progesterone, pregnenolone, dehydroepiandrosterone, desoxycorticosterone, 3 alpha,5 alpha-tetrahydrodesoxycorticosterone, 3 alpha-hydroxy-5 alpha-dihydroprogesterone, 17-hydroxyprogesterone, and 17-hydroxypregnenolone together with their esterified forms. As pregnenolone is the precursor of all steroids, its formation was studied in detail as an index of a steroid-synthesizing tissue. Pregnenolon…

Human neuroblastoma SH-SY5Y cell line: neurosteroid-producing cell line relying on cytoskeletal organization.

2000

Pregnenolone, the precursor of all steroids, is synthesized by CNS structures. The synthesis requires an obligatory step involving cholesterol transport to mitochondrial cytochrome P450-cholesterol side chain cleavage (cytP450scc), although the underlying mechanism(s) are still mostly unknown. We used the human neuroblastoma SH-SY5Y cell line to investigate cytP450scc expression and activity and to establish a role of cytoskeleton in pregnenolone synthesis. Immunocytochemical and biochemical approaches revealed that undifferentiated as well as differentiated cells either by retinoic acid (RA) or phorbol ester 12-O-tetradecanoylphorbol-13-acetate (TPA), possess cytP450scc and rapidly synthes…

The climate change effect on the african monsoon region : evolution of the precipitation and atmospheric fields in the CMIP3 and CMIP5 simulations un…

2013

The fourth IPCC report in 2007 established the synthesis of previously published work onthe effects of climate change on global and regional scales. It shows in particular that no consensuscan be found with regard to the future of rainfall — and atmospheric dynamics- associatedwith region — African monsoon. This dissertation revisits this issue in the light of new dataand using an approach avoiding over-representation of the number of simulations available forone type of model and taking into account the diversity of models and their evolution in time :twenty general circulation models (GCMs) participating in the exercises CMIP3 (twelve GCMs)and CMIP5 (eight GCMs) under the A1B emissions sc…

Comparative Analyses for Gluten-Free Biscuits from Mixture Oat and Corn

2018

Abstract Gluten-free products are necessary for the people suffering from celiac disease or having a sensibility for products containing gluten. The aim of this study was to prepare and evaluate gluten free biscuits made from a mixture of flours (50% corn flour and 50% oat flour) with various fats (palm fat, coconut oil) and sweeteners (sugar, stevioside with erythritol). Sample biscuits were made and then analyzed to find out the moisture, water activity, braking point, specific volume and sensory evaluation. All samples had the moisture close to 5-6%. Water activity ranged from 0.49 to 0.71. The breaking point was higher for sample prepared with palm fat and sugar (0.43). This sample was …

Determination of major human cytochrome P450s activities in 96-well plates using liquid chromatography tandem mass spectrometry.

2007

At the early stage of drug discovery, thousands of new chemical entities (NCEs) may be screened before a single candidate can be identified for development. Evaluation of the effect of NCEs on human CYP450 enzyme activities is a key issue in pharmaceutical development as it may explain inter-subject variability, drug-drug interactions, non-linear pharmacokinetics and toxic effects. A liquid chromatography tandem mass spectrometry (HPLC-MS/MS) method has been developed for the fast and routine analysis of major human CYP450s enzyme activities (CYP1A2, CYP2A6, CYP2B6, CYP2C9, CYP2C19, CYP2D6, CYP2E1 and CYP3A4) in primary hepatocyte cell cultures. The high sensitivity and selectivity of mass …
