Background: Various trivalent radiometals are well suited for labeling of DOTA-conjugated variants of Glu-ureido-based prostate-specific membrane antigen (PSMA) inhibitors. The DOTA-conjugate PSMA-617 has proven high potential in PSMA radioligand therapy (PSMA-RLT) of prostate cancer as well as PET imaging when labeled with lutetium-177 and gallium-68 respectively. Considering the relatively short physical half-life of gallium-68 this positron emitter precludes prolonged acquisition periods, as required for pre-therapeutic dosimetry or intraoperative applications. Luting of ceramic crowns with a self-adhesive cement: Effect of contamination on marginal adaptation and fracture strength

Objectives: This study evaluated the percentages of continuous margins (%CM) and fracture strength (FS) of crowns made out from blocs of leucite-reinforced ceramic (IPS Empress CAD) and luted with a representative self-adhesive cement (RelyX Unicem) under four contaminating agents: saliva, water, blood, a haemostatic solution containing aluminium chloride (pH= 0.8) and a control group with no contamination. Study Design: %CM at both tooth-cement (TC) and cement-crown (CC) interfaces were determined before and after a fatigue test consisting of 600’000 chewing loads and 1’500 temperature cycles changing from 5º C to 50º C. Wild-type JAK2 secondary acute erythroleukemia developing after JAK2-V617F-mutated primary myelofibrosis.

A 54-year-old female patient developed acute erythroleukemia after an 8-year course of primary myelofibrosis. The latter harbors the JAK2-V617F mutation and was treated with hydroxyurea and anagrelide. A bone marrow trephine biopsy disclosed 2 morphologically distinct areas of chronic primary myelofibrosis and acute erythroleukemia. Microdissection and a separate molecular pathological analysis was performed. Although the activating JAK2-V617F mutation was not maintained in blasts of acute erythroleukemia, it was detectable in the chronic phase of primary myelofibrosis, indicating that this mutation did not play a role in the leukemic transformation of erythroid cells.

Mechanical ventilation and Streptococcus pneumoniae pneumonia alter mitochondrial homeostasis.

Abstract Required mechanical ventilation (MV) may contribute to bacterial dissemination in patients with Streptococcus pneumoniae pneumonia. Significant variations in plasma mitochondrial DNA (mtDNA) have been reported in sepsis according to the outcome. The impact of lung stretch during MV was addressed in a model of pneumonia. Healthy or S. pneumoniae infected rabbits were submitted to MV or kept spontaneously breathing (SB). Bacterial burden, cytokines release, mitochondrial DNA levels, integrity and transcription were assessed along with 48-hour mortality.
