MiR-29b-1 expression impaired Cancer Stem-Like properties of human osteosarcoma 3AB-OS cells in vitro.

2013

Osteosarcoma (OS) is the most common type of bone cancer, with a peak incidence in the early childhood. Emerging evidence suggests that treatments targeting cancer stem cells (CSCs) within a tumor can halt cancer and improve patient survival. Although microRNAs are frequently dysregulated in human cancers, if they influence OS malignancy and whether or not targeting CSC-associated microRNAs inhibit OS progression remain unclear. Recently (1), we described a predictive network for two downregulated miRNA family (let-7/98 and miR-29a,b,c) and their upregulated anticorrelated mRNAs. Here, we investigated in vitro the role of miR-29b-1 in regulating cell proliferation, clonogenic growth and che…

MicroRNA-29b-1 is involved in self-renewal and fate decisions of human osteosarcoma 3AB-OS cancer stem cells

2014

Emerging evidence suggests that treatments targeting cancer stem cells (CSCs) within a tumor can halt cancer and improve patient survival. Moreover, identification of CSC-related MicroRNAs (miRNAs) would provide information for a better understanding of CSCs. miR-29 family is a class of miRNAs aberrantly expressed in multiple cancers. They are frequently down-regulated in osteosarcoma (OS), the most common form of childhood cancer with a potent metastasizing potential. 3AB-OS CSC, a human pluripotent CSC line by us produced from the human osteosarcoma MG63 cells (1) is a useful model to study CSC origin and roles (2). Previously, we have shown that in 3AB-OS CSCs miR-29b is potently down-re…

The X-ray emission of the supernova remnant W49B observed withXMM-Newton

2006

In the framework of the study of supernova remnants and their complex interaction with the interstellar medium, we report on an XMM-Newton EPIC observation of the Galactic supernova remnant W49B. We investigate the spatial distribution of the chemical and physical properties of the plasma, so as to get important constraints on the physical scenario, on the dynamics of the supernova explosion, and on the interaction of the supernova remnant with the ambient interstellar clouds. We present line images, equivalent width maps, and a spatially resolved spectral analysis of a set of homogeneous regions. The X-ray spectrum of W49B is characterized by strong K emission lines from Si, S, Ar, Ca and …

Off-Axis Properties of Short Gamma-Ray Bursts

2005

Based on recent models of relativistic jet formation by thermal energy deposition around black hole-torus systems, the relation between the on- and off-axis appearance of short, hard gamma-ray bursts (GRBs) is discussed in terms of energetics, duration, average Lorentz factor, and probability of observation, assuming that the central engines are remnants of binary neutron star or neutron star-black hole mergers. As a consequence of the interaction with the torus matter at the jet basis and the subsequent expansion of the jets into an extremely low-density environment, the collimated ultrarelativistic outflows possess flat core profiles with only little variation of radially-averaged propert…

miR-29s: A family of epi-miRNAs with therapeutic implications in hematologic malignancies

2015

A wealth of studies has highlighted the biological complexity of hematologic malignancies and the role of dysregulated signal transduction pathways. Along with the crucial role of genetic abnormalities, epigenetic aberrations are nowadays emerging as relevant players in cancer development, and significant research efforts are currently focusing on mechanisms by which histone post-translational modifications, DNA methylation and noncoding RNAs contribute to the pathobiology of cancer. As a consequence, these studies have provided the rationale for the development of epigenetic drugs, such as histone deacetylase inhibitors and demethylating compounds, some of which are currently in advanced p…

Role of sparc and MIR-29B1 in molecular effects induced by win in osteosarcoma MG63 cells

2014

SPARC (Secreted protein acidic and rich in cysteine) is considered as a prototype of matricellular protein due to its structure and the function that it displays in regulating cell/extracellular microenvironment interactions during development and in response to injury. Earlier studies underlined pleiotropic effects of intracellular SPARC on cancer growth and, in some cancer cell lines, identified it as a tumor suppressor protein. Objective This study aimed to evaluate the role of SPARC and its related miRNA in the molecular effects induced by the cannabinoid WIN in osteosarcoma MG63 cells. In these cells WIN is not able to induce cell death but sensitizes cells to TRAIL-mediated apoptotic …
