Tyrosine-phosphorylation-dependent and Rho-protein-mediated control of cellular phosphatidylinositol 4,5-bisphosphate levels

1998

The polyphosphoinositide PtdIns(4,5)P2, best known as a substrate for phospholipase C isozymes, has recently been recognized to be involved in a variety of other cellular processes. The aim of this study was to examine whether the cellular levels of this versatile phospholipid are controlled by tyrosine phosphorylation. The studies were performed in human embryonic kidney (HEK)-293 cells stably expressing the M3 muscarinic acetylcholine receptor. Inhibition of tyrosine phosphatases by pervanadate induced an up-to-approx.-2.5-fold increase in the total cellular level of PtdIns(4,5)P2, which was both time- and concentration-dependent. In contrast, the tyrosine kinase inhibitors, genistein and…

Restoration of Clostridium difficile toxin-B-inhibited phospholipase D by phosphatidylinositol 4,5-bisphosphate.

1996

Receptor signalling to phospholipase D (PLD) in human embryonic kidney (HEK) cells stably expressing the m3 muscarinic acetylcholine receptor apparently involves Rho proteins. Since phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P2] has been recognized as an essential cofactor for PLD activity and since activated Rho proteins have been reported to stimulate the synthesis of PtdIns(4,5)P2, we studied whether in HEK cells PLD activity is regulated by PtdIns(4,5)P2 and, in particular, whether PtdIns(4,5)P2 can restore PLD activity inhibited by Clostridium difficile toxin B, which inactivates Rho proteins. Addition of MgATP to permeabilized HEK cells increased basal PLD activity and potentia…

Complement and Atherogenesis

1999

Abstract —Complement activation occurs in temporal correlation with the subendothelial deposition of LDL during early atherogenesis, and complement also plays a pathogenetic role in promoting lesion progression. Two lesion components have been identified that may be responsible for complement activation. First, enzymatic degradation of LDL generates a derivative that can spontaneously activate complement, and enzymatically degraded LDL (E-LDL) has been detected in the lesions. Second, C-reactive protein (CRP) colocalizes with complement C5b-9, as evidenced by immunohistological studies of early atherosclerotic lesions, so the possibility exists that this acute phase protein also fulfills a…

DNA-binding and in vitro cytotoxic activity of platinum(II) complexes of curcumin and caffeine

2019

Abstract Three Pt(II) complexes containing the natural ligands curcumin and caffeine, namely [Pt(curc)(PPh3)2]Cl (1), [PtCl(curc)(DMSO)] (2) (curc = deprotonated curcumin) and trans-[Pt(caffeine)Cl2(DMSO)] (3), were synthesized and fully characterized. The data obtained suggest that, for both 1 and 2, the anion of curcumin is coordinated to the platinum ion via the oxygen atoms of the β-diketonate moiety. Spectroscopic features reveal that in 2 and 3, a DMSO molecule is S-bonded to the metal centre. For 3, all data indicate a square-planar geometry formed by a 9-N bonded caffeine, two trans chloride anions and a DMSO. The three complexes undergo changes in solution upon incubation for 24 h;…

Short-lived luminescence of mixed silver halides

1993

Abstract Two kinds of excitonic luminescence were investigated for the entire series of solid solutions, AgBr1−xClx, (0 ⪕ x ⪕ 1). The time-resolved spectra observed around 2.5 eV after both pulsed high-power electron beam and N2-laser irradiations are analyzed in terms of the diffusion-controlled recombination of Frenkel defects Ag0i and Ag2+V-c. It is shown that the effective activation energy of Ag0i migration is a linear function of the composition x; and predominantly two kinds of nearest neighbours contribute to this process. The position of maximum of the excitonic molecule luminescence found in mixed monocrystals and fibers reveals a distinctive dependence on the composition x. Its a…

Temperature dependence of luminescence decay in Sn-doped silica

2005

We report an experimental study on the temperature dependence, in the range 18-300 K, of the decay kinetics of the emission at 4.1 eV from the first excited electronic state of oxygen deficient centers in a 2000 ppm Sn-doped sol-gel silica. At low temperature, this luminescence decays exponentially with a lifetime of 8.4 ns, whereas, on increasing the temperature, the time decay decreases and cannot be fitted with an exponential function. These results are expected if there is a competition between the radiative and the thermally activated intersystem-crossing decay channels toward the associated triplet state. The comparison with previous data in pure oxygen-deficient and Ge-doped silica g…

Conformational disorder and optical properties of point defects in vitreous silica

2004

Abstract Disordered systems are characterized by the presence of local conformational heterogeneity, which reflects the complex landscape of the potential energy of the vitreous state. Optical properties of defects embedded in a vitreous matrix are also determined by the interaction with the surrounding environment; so the conformational disorder of the system induces spectral inhomogeneity. As a consequence, detailed experimental investigation of absorption and photoluminescence bands can give information on configurational substates around the chromophore. We focused our attention on B-type optical activity in silica glasses, characterized by a singlet emission and a triplet emission, conne…

Temperature dependence of O2 singlet photoluminescence in silica nanoparticles

2013

Abstract The near infrared singlet emission and photoluminescence lifetime of O 2 molecules embedded in silica nanoparticles are studied from room temperature down to 10 K. The area of the photoluminescence band under infrared excitation decreases for temperature above 100 K and the lifetime is shortened. These observations provide evidence of a thermally activated relaxation channel with activation energy of about 40 meV. This relaxation mechanism adds to the already known temperature independent electronic-to-vibrational coupling involving high energy vibrational modes of the host matrix or its impurities. The thermally activated process is suggested to consist in the breakage of the O 2 …

Exchange between interstitial oxygen molecules and network oxygen atoms in amorphousSiO2studied byO18isotope labeling and infrared photoluminescence …

2011

Amorphous ${\mathrm{SiO}}_{2}$ ($a$-${\mathrm{SiO}}_{2}$) thermally annealed in an oxygen atmosphere incorporates oxygen molecules (O${}_{2}$) in interstitial voids. When the thermal annealing is performed in $^{18}\mathrm{O}$${}_{2}$ gas, interstitial $^{18}\mathrm{O}$${}_{2}$ as well as interstitial $^{16}\mathrm{O}$$^{18}\mathrm{O}$ and $^{16}\mathrm{O}$${}_{2}$ are formed due to the oxygen exchange with the $a$-${\mathrm{SiO}}_{2}$ network. The ${a}^{1}{\ensuremath{\Delta}}_{g}(v=0)\ensuremath{\rightarrow}{X}^{3}{\ensuremath{\Sigma}}_{g}^{\ensuremath{-}}(v=1)$ infrared photoluminescence band of interstitial ${\mathrm{O}}_{2}$ was utilized to quantitatively analyze the oxygen exchange, t…
