Background & Aims Serum microRNA (miRNA) levels are known to change in non-alcoholic fatty liver disease (NAFLD) and may serve as useful biomarkers. This study aimed to profile miRNAs comprehensively at all NAFLD stages. Methods We profiled 2,083 serum miRNAs in a discovery cohort (183 cases with NAFLD representing the complete NAFLD spectrum and 10 population controls). miRNA libraries generated by HTG EdgeSeq were sequenced by Illumina NextSeq. A transmembrane serine residue in the Rot1 protein is essential for yeast cell viability

Polar residues are present in TM (transmembrane) helices and may influence the folding or association of membrane proteins. In the present study, we use an in vivo approach to analyse the functional and structural roles for amino acids in membrane-spanning motifs using the Rot1 (reversal of Tor2 lethality 1) protein as a model. Rot1 is an essential membrane protein in Saccharomyces cerevisiae and it contains a single TM domain. An alanine insertion scanning analysis of this TM helix revealed that the integrity of the central domain is essential for protein function. Sepiapterin reductase in cultured human cells.

Sepiapterin reductase, an enzyme involved in the synthesis of tetrahydrobiopterin (the natural cofactor for phenylalanine, tyrosine and tryptophan hydroxylases), has been assayed in cultured human amniotic fibroblasts and in cultured mononuclear blood cells. In both cases, the Michaelis constants for sepiapterin and NADPH were essentially equal; 20 microM and 6 microM respectively for stimulated mononuclear blood cells and 22 microM and 5 microM respectively for amniotic fibroblasts. The inhibition by N-acetylserotonin was also similar in both cases. The concentration that produced 50% inhibition in stimulated mononuclear blood cells and in amniotic fibroblasts was 2 microM. We have studied the ESR response of alanine pellets with and without gadolinium exposed to the thermal column of the TRIGA Mark II research reactor at the University of Mainz (Germany). The choice of Gd as additive nucleus is due to its very high capture cross section to thermal neutrons and to the possibility for secondary particles produced after interaction with thermal neutrons of releasing their energy in the neighborhood of the reaction site. In particular, it was found that low concentration (5% by weight) of Gd brings about a neutron sensitivity enhancement of more than 10 times without heavily reducing tissue equivalence. The aminoacid L-a-alanine has attracted considerable interest for use in radiation ESR dosimetry and has been formally accepted as a secondary standard for high-dose (kGy) and transfer dosimetry .
