Water, sodium, potassium and chloride balances, food uptake and changes in body weight were determined in large groups of adrenalectomized or sham-operated rats having permanent access to water, 0.9 per cent saline and rat crackers. Summary The data reported in the present paper refer to quantitative variations of plasma renin activity and plasma aldosterone levels after administration of alpha-blocking agents, i.e. phentolamine, phenoxybenzamine and gihydroergotoxin derivatives, either to rats kept on a standard diet with water ad libitum or to rats receiving distilled water load, the latter treatment causing an increase of both plasma renin activity and plasma aldosterone levels if compared to control values. Abstract The data reported in the present paper refer to quantitative variations of plasma renin activity (PRA.) and of plasma aldosterone levels in rats after adiministration of saline, chrystalloid and colloid solutions. In our experimental conditions PRA and plasma aldosterone levels were shown to increase significantly after administration of distilled water and glucose solution. Variations of PRA and plasma aldosterone levels were only slight when plasma volume was made to increase due to administration of polyvinylpirrolidone solution. PRA, instead, was significantly lower in rats having received saline salution.

New derivatives of progesterone and aldosterone were synthesized and functionally tested with commercially available antibodies. The covalent labelling of antibodies specific for aldosterone and progesterone was detected by SDS/PAGE analysis and subsequent autoradiography after using 3-(O-carboxymethyl)-oximino-(3-[125I]iodo-4-azidosalicylamidobu tylamine) derivatives of aldosterone and progesterone, respectively, as photoactivatable radioligands. In experimental studies conducted in rats, raising serum uric acid (SUA) levels resulted in stimulation of intrarenal renin expression. Studies in humans exploring the association of SUA with plasma renin activity (PRA) yielded conflicting results. The 20 alpha-reduced derivative of aldosterone, 20 alpha-dihydroaldosterone, was needed as reference compound in order to continue the studies on 18-hydroxylation in the Y-1 adrenal cell line. It was obtained by reduction of aldosterone with sodium borohydride. Analysis of the products of the reaction as methoxime trimethylsilyl (MO-TMS) derivatives by gas chromatography (GC) and GC-mass spectrometry (GC-MS) showed three possible forms of the compound. Their identification was confirmed by comparison with the products obtained by stereospecific reduction of aldosterone using 3 alpha,20 beta-hydroxysteroid dehydrogenase.
