Dihydrooxazine Oxides as Key Intermediates in Organocatalytic Michael Additions of Aldehydes to Nitroalkenes

Pause and play: dihydrooxazine oxides are stable intermediates that are protonated directly, without the intermediacy of the zwitterions, in organocatalytic Michael additions of aldehydes and nitroalkenes (see scheme, R=alkyl). Protonation of these species explains both the role of the acid co-catalyst in these reactions, and the observed stereochemistry when the reaction is conducted with α-alkylnitroalkenes.

Mixed alkyl-methyl- and aryl-methylorganoaluminum chlorides 6 were formed by reaction of methylaluminum dichloride with organolithium or Grignard compounds and used for chemoselective 1,4 addition of higher alkyl, aryl, alkenyl and alkinyl groups to alkylidine malonic esters 1 and 2. As an alternative, mixed trimethylorganoaluminates 7 can also be applied for these Michael addition reactions. For conjugate addition of alkenyl groups to alkylidene malonates 1 and 2, alkenyl diisopropylalanes 10 obtained from alkynes and diisopropylaluminum hydride proved the most efficient reagents. 4-(1-Cycloalken-1-yl)-1,2-dialkoxybenzenes 8, 10, and 11 and 5-(1-cycloalken-1-yl)-1,3-benzodioxoles 12−15 react with the semisquaric halides 5a and 5b in a dehydrative annulation process to give the annulated α-halobenzocyclobutenones 9a,b and 16a,b−21a,b in poor to good yields (20−76%). The reaction failed with alicyclic−aromatic dienes having no or only one alkoxy group in the benzene ring. The dehydrative annulation process could be extended to 4-(3,4-dimethoxyphenyl)-1,2-dihydronaphthalene (24), affording the highly annulated α-chlorobenzocyclobutenone 25 in 50% yield. The effect of various aromatic and aliphatic dielectrophiles on the lithium azaenolates of 2-alkyl-2-oxazolines and 2-alkyl-2-thiazolines has been examined. This effect varies greatly, depending on the nature of the dielectrophile used. 3-Aminoindene (3) and 3-alkylidenephthalimidine (4−5) derivatives were formed as a result of the reactions with dielectrophiles derived from ortho-substituted benzonitriles. Similarly, 2-alkylidenepyrrolidine (7) and 2-alkylidenepiperidine (8) derivatives were obtained in high yields from 2-alkyl-2-oxazolines or 2-alkyl-2-thiazolines and aliphatic dielectrophiles derived from ω-haloalkyldiphenylacetonitrile. C−C versus C−N annulation reactions are discussed.

Novel Porphyrin-Cholic Acid Conjugates as Receptors for Biologically Important Anions

A series of novel receptors showing high binding affinity in aqueous media for biologically important anions are reported. These naturally chromophoric porphyrin-based receptors contain cholic acids connected via quaternary alkyl ammonium amido linkages.
