A novel bio-orthogonal cross-linker for improved protein/protein interaction analysis

International audience; The variety of protein cross-linkers developed in recent years illustrates the current requirement for efficient reagents optimized for mass spectrometry (MS) analysis. To date, the most widely used strategy relies on commercial cross-linkers that bear an isotopically labeled tag and N-hydroxysuccinimid-ester (NHS-ester) moieties. Moreover, an enrichment step using liquid chromatography is usually performed after enzymatic digestion of the cross-linked proteins. Unfortunately, this approach suffers from several limitations. Energy-energy correlations in hadronic final states from Z0 decays

We have studied the energy-energy angular correlations in hadronic final states from Z0 decay using the DELPHI detector at LEP. From a comparison with Monte Carlo calculations based on the exact second order QCD matrix element and string fragmentation we find that Λ(5)/MS = 104-20 +25 (stat.)-20 +25(syst.)-00 +30(theor.) MeV, which corresponds to αs(91 GeV) = 0.106± 0.003 (stat.)±0.003(syst.)-0.000 +0.003(theor.). The theoretical error stems from different choices for the renormalization scale of αs. In the Monte Carlo simulation the scale of αs as well as the fragmentation parameters have been optimized to described reasonably well all aspects of multihadron production.

Alpha-synuclein is a protein of 140 amino acids, intrinsically disordered and found abundantly in the brain and whose toxic aggregation there is the hallmark of the Parkinson's disease.The aim of the thesis is to understand the effects of certain mutations on the dimerization of alpha-synuclein and to contribute to understand its involvement in Parkinson's disease by using coarse-grained molecular dynamics. Influence of ADAM10 on prion protein processing and scrapie infectiosity in vivo.

Abstract Both the cellular prion protein (PrPc) and the amyloid precursor protein (APP) are physiologically subjected to complex proteolytic processing events. While for APP the proteinases involved – alpha-, beta- and gamma-secretase – have been identified in vitro and in vivo, the cleavage of PrPc by now has been linked only to the shedding activity of the metalloproteinase ADAM10 and/or ADAM17 in cell culture. Here we show that neuronal overexpression of the alpha-secretase ADAM10 in mice reduces all PrPc species detected in the brain instead of leading to enhanced amounts of specific cleavage products of PrPc. Energy dependence of the differences between the quark and gluon jet fragmentation

Three jet events arising from decays of the Z beson, collected by the DELPHI detector, were used to measure differences in quark and gluon fragmentation. Gluon jets were anti-tagged by identifying b quark jets. Unbiased quark jets came from events with two jets plus one photon. Quark and gluon jet properties in different energy ranges were compared for the first time within the same detector. Quark and gluon jets of nearly the same energy in symmetric three jet event topologies were also compared. Vulnerability of peripheral catecholaminergic neurons to MPTP is not regulated by alpha-synuclein.

Although generally considered a prototypical movement disorder, Parkinson's disease is commonly associated with a broad-spectrum of non-motor symptoms, including autonomic dysfunctions caused by significant alterations in catecholaminergic neurons of the peripheral sympathetic nervous system. Here we present evidence that alpha-synuclein is highly expressed by sympathetic ganglion neurons throughout embryonic and postnatal life and that it is found in tyrosine hydroxylase-positive sympathetic fibers innervating the heart of adult mice. α-Synuclein (α-syn) is a small protein of unknown function that is found aggregated in Lewy bodies, the histopathological hallmark of sporadic Parkinson disease and other synucleinopathies. Mutations in the α-syn gene and a triplication of its gene locus have been identified in early onset familial Parkinson disease. α-Syn turnover can be mediated by the proteasome pathway. It is based on a covariation pseudo-additivity and Morse-Transue's integral with respect to a bimesure built by using pseudo-additivity property.
