Cellular stress induces cap-independent alpha-enolase/MBP-1 translation.

AbstractMyc promoter-binding protein-1 (MBP-1) is a shorter protein variant of the glycolytic enzyme alpha-enolase. Although several lines of evidence indicate that MBP-1 acts as a tumor suppressor, the cellular mechanisms and signaling pathways underlying MBP-1 expression still remain largely elusive. To dissect these pathways, we used the SkBr3 breast cancer cell line and non-tumorigenic HEK293T cells ectopically overexpressing alpha-enolase/MBP-1. The role of alpha-synuclein (alpha-Syn) has recently received considerable attention because it seems to play a role in Parkinson's disease (PD). Missense mutations in the alpha-Syn gene were found in autosomal dominant PD and alpha-Syn was shown to be a major constituent of protein aggregates in sporadic PD and other synucleinopathies. Tässä tutkielmassa käsittelen Monte Carlo -puuhakualgoritmia sekä sen roolia hyvin menestyneessä AlphaGo-tekoälyssä. Tavoitteena oli muodostaa kokonaiskuva AlphaGo:n toiminnasta, painottuen etenkin Monte Carlo -puuhaun näkökulmaan. Tutkimuksen perusteella selvisi miten ohjelma hyödyntää omassa hakualgoritmissään kyseistä puuhakua, jota se parantaa hyödyntäen koneoppimista ja useita eri tarkoituksiin opetettuja neuroverkkoja. In the milk of ruminants, more than 95% of proteins are synthesized by six structural genes, four caseins (alphas1, beta, alphas2 and κ-caseins) and two whey proteins (alpha-lactalbumin and beta-lactoglobulin). Goat casein genes have been found to show high polymorphism, which influences not only the quantity of casein in milk but also the structural and nutritional characteristics and technological properties of milk. Biomarkers Related to Synaptic Dysfunction to Discriminate Alzheimer’s Disease from Other Neurological Disorders

Recently, the synaptic proteins neurogranin (Ng) and α-synuclein (α-Syn) have attracted scientific interest as potential biomarkers for synaptic dysfunction in neurodegenerative diseases. In this study, we measured the CSF Ng and α-Syn concentrations in patients affected by AD (n = 69), non-AD neurodegenerative disorders (n-AD = 50) and non-degenerative disorders (n-ND, n = 98). The concentrations of CSF Ng and α-Syn were significantly higher in AD than in n-AD and n-ND. Faldaprevir (BI 201335), BI 207127 and ribavirin oral therapy for treatment-naive HCV genotype 1: SOUND-C1 final results

Background Faldaprevir (BI 201335) and deleobuvir (BI 207127) are direct-acting antiviral agents under development for the treatment of chronic HCV infection. This article describes the final results of the Phase Ib SOUND-C1 study that evaluated the interferon-free oral combination of faldaprevir, deleobuvir and ribavirin in 32 treatment-naive patients infected with HCV genotype 1. Methods Patients were randomized to receive deleobuvir 400 mg ( n=15) or 600 mg ( n=17) three times daily plus faldaprevir 120 mg once daily and weight-based ribavirin for 4 weeks. International audience; Using ultra performance liquid chromatography-mass spectrometry we identified for the first time glutamine-N-alpha-conjugates of the fatty acids (E)/(Z)-3-methylhex-2-enoic acid and (R)/(S)-3-hydroxy-3-methylhexanoic acid as well as cysteinylglycine-S-conjugates of (R)/(S)-3-methyl-3-sulphanylhexan-1-ol and (R)/(S)-3-sulphanylhexan-1-ol as constituents of human milk and colostrum. The glutamine-N-alpha-conjugates were detected also in human amniotic fluids. AbstractOligomeric species formed during α-synuclein fibrillation are suggested to be membrane-disrupting agents, and have been associated with cytotoxicity in Parkinson’s disease. The majority of studies, however, have revealed that the effect of α-synuclein oligomers is only noticeable on systems composed of anionic lipids, while the more physiologically relevant zwitterionic lipids remain intact. We present experimental evidence for significant morphological changes in zwitterionic membranes containing cholesterol, induced by α-synuclein oligomers.
