Two changes of the same nucleotide confer resistance to diuron and antimycin in the mitochondrial cytochrome b gene of Schizosaccharomyces pombe

1988

AbstractDiuron (3-(3,4-dichlorophenyl)-1,1-dimethylurea) and antimycin, both inhibitors of mitochondrial respiration, block electron flow between cytochromes b and c1. Mutants resistant to either drug have been selected using Schizosaccharomyces pombe strains with an extrachromosomally inherited mutator. In analogy to Saccharomyces cerevisiae these mutational sites were assumed to map in the cytochrome b gene. DNA sequence analysis showed that two changes in the same nucleotide are responsible for resistance to antimycin and diuron. Analysis of resistant and sensitive progeny of crosses between the mutants and the wild type confirmed the correlation between mutational alteration and resista…

Dental pulp in mature human teeth: morphological alterations and metalloproteinases-2 and -9, Annexin-5, BCL-2, and iNOS modulation.

2015

Tooth replantation, as a treatment concept, has been subject to controversies regarding the mechanism as well as the various parameters underlying this process. This work aimed to study time-related changes in the pulp of replanted mature human premolars through the changes in the levels of certain factors involved in the underlying mechanisms of pulpal tissue healing after replantation. Eleven experimental mature teeth were extracted, immediately replanted in the original socket and left without any other intervention for 1, 2, 3 and 12 weeks before re-extraction. Three premolars served as control. All specimens were subject to histological analysis and the levels of MMP-2, MMP-9, Annexin …

RB, epigenetic changes and chromosomal alterations in human primary fibroblasts in culture

2008

The regulation of chromatin structure is a dynamic and complex process modulated by epigenetic mechanisms. Epigenetic changes as malfunctioning of histone modifications and DNA methylation could affect several different cellular processes like regulation of gene transcription and could compromise the correct chromosome condensation and segregation. Is important to note that these alterations have been correlated with cancer initiation/progression. In particular hypomethylation of pericentromeric regions, usually methylated, has been associated to chromosomal instability, as well as hypermethylation of promoter CpG islands of tumor suppressor genes (p16, CHFR, BRCA1) is considered a cause of…

CHARACTERIZATION OF TRANSFORMED CELL LINES OBTAINED FROM PRIMARY RAT CORTICAL ASTROCYTES

2021

Brain cancers are complex and heterogeneous; most of them derive from glial cells[1], and are called gliomas, further subdivided into astrocytomas, oligodendrogliomas, ependymomas and glioastrocytomas[2]. The malignant cells undergo modifications of their metabolism and behaviour, and acquire the ability to migrate along the blood vessels in small groups (model of the guerrilla war)[3], thus invading the surrounding brain parenchyma. Most important, they have the capacity to affect the surrounding microenvironment, by altering both the extracellular matrix and the properties of the normal cells present in the brain, including glial-, endothelial-, and immune-cells, further promoting cancer …

An In Vitro Model of Glioma Development

2023

Gliomas are the prevalent forms of brain cancer and derive from glial cells. Among them, astrocytomas are the most frequent. Astrocytes are fundamental for most brain functions, as they contribute to neuronal metabolism and neurotransmission. When they acquire cancer properties, their functions are altered, and, in addition, they start invading the brain parenchyma. Thus, a better knowledge of transformed astrocyte molecular properties is essential. With this aim, we previously developed rat astrocyte clones with increasing cancer properties. In this study, we used proteomic analysis to compare the most transformed clone (A-FC6) with normal primary astrocytes. We found that 154 proteins are…

Summary ― The work presents the results of the bio-archaeological analyzes carried out on human skeletal remains found in fourteen burials of Medieval chronology at the Archaeological Park of Valle dei Templi in Agrigentum (Sicily). The aim of the work was the acquisition of the information necessary for the reconstruction of the biological profile of each individual throught the determination of sex the estimate of the biological age at death the estimate of height and the evaluation of pathologies and markers of occupational stress through current anthropological diagnostic methodologies and techniques. Althought the poor state of conservation of some individuals has not allowed their whole anthropological characterization the investigations have highlighted the heterogeneity relating to the age groups and have allowed us to ascertain the presence of pathological alterations in subjects of mature age sometimes of unknown etiology such as DISH (Diffuse Idiopathic Hyperostosis). The sample size is not representative of the entire population but subsequent insights will provide a better understanding of the population dynamics of Medieval Agrigentum.Settore BIO/08 - Antropologia

Raman-Mössbauer-XRD studies of selected samples from “Los Azulejos” outcrop: A possible analogue for assessing the alteration processes on Mars

2016

The outcrop of “Los Azulejos” is visible at the interior of the Cañadas Caldera in Tenerife Island (Spain). It exhibits a great variety of alteration processes that could be considered as terrestrial analogue for several geological processes on Mars. This outcrop is particularly interesting due to the content of clays, zeolite, iron oxides, and sulfates corresponding to a hydrothermal alteration catalogued as “Azulejos” type alteration. A detailed analysis by portable and laboratory Raman systems as well as other different techniques such as X ray diffraction (XRD) and Mössbauer spectroscopy has been carried out (using twin-instruments from Martian lander missions: Mössbauer spectrometer MI…

The role of Mediterranean wetlands in climate regulation and greenhouse gases exchanges: from carbon biogeochemistry to European water and nature Dir…

2022

Monthly Hydrological Indicators to Assess Possible Alterations on Rivers’ Flow Regime

2018

Assessing potential deviations of the fundamental river basins’ hydrological processes and streamflow characteristics from the “natural trajectory” represents a high-priority objective to understand the biological impact of altered flow regime on river ecosystems. Existing approaches are mainly based on the analysis of daily-based indicators of hydrologic alteration, which requires wide database, including “pre-impact” and “post-impact” daily flow data frequently unavailable. The hydrological modeling is commonly used to face data missing problems or reconstruct natural conditions, even if models, especially at the daily scales, are often complex and computationally intensive. The use of si…

Advance in Vascular Phenotype Assessment in Children and Adolescents

2010

Cardiovascular damage occurring in adults finds its roots in risk factors operating early in life. Among the factors influencing cardiovascular risk, blood pressure values represent an important measurable marker of the level of potential cardiovascular risk in children and adolescents. Indeed, there is growing evidence that mild blood pressure elevations are much more common than was thought in the past in a pediatric population. Furthermore, hypertension in childhood has gained ground in cardiovascular medicine thanks to the progress made in several areas of pathophysiological and clinical research.1 It is not uncommon for high blood pressure in the young to be accompanied by evidence of …
