Enzymatic Spermine Metabolites Induce Apoptosis Associated with Increase of p53, caspase-3 and miR-34a in Both Neuroblastoma Cells, SJNKP and the N-M…

2021

Neuroblastoma (NB) is a common malignant solid tumor in children and accounts for 15% of childhood cancer mortality. Amplification of the N-Myc oncogene is a well-established poor prognostic marker in NB patients and strongly correlates with higher tumor aggression and resistance to treatment. New therapies for patients with N-Myc-amplified NB need to be developed. After treating NB cells with BSAO/SPM, the detection of apoptosis was determined after annexin V-FITC labeling and DNA staining with propidium iodide. The mitochondrial membrane potential activity was checked, labeling cells with the probe JC-1 dye. We analyzed, by real-time RT-PCR, the transcript of genes involved in the apoptot…

Don't change a winning horse

2005

IntroductionThere is surprising excitement about the adequacy andfuture of the somatoform disorders (SFDs) as a circum-scribed diagnostic group [1–4]. Some authors wish to takethe forthcoming fifth revision of the Diagnostic andStatistical Manual (DSM) as a welcome occasion to removean unloved diagnostic term. However, eliminating the SFDswould devaluate the progress of research and clinicalpractice made during the past 25 years. It would lead toinsecurity and confusion in an interdisciplinary field whereclear terminology and common understanding is of highvalue. This contribution attempts to summarize some of themajor reasons why DSM-V should continue with the SFDs.Some suggestions will be…

Survey of five major grapevine viruses infecting Blatina and Žilavka cultivars in Bosnia and Herzegovina

2021

The sanitary status of grapevines has not yet been considered sufficiently in vineyards throughout Bosnia and Herzegovina (BiH). An extensive survey of five major grapevine viruses in the country was carried out in 2019. A total of 630 samples from the two dominant autochthonous cultivars, named Žilavka and Blatina, were tested by DAS-ELISA for the presence of grapevine leafroll-associated viruses (GLRaV-1 and 3), grapevine fleck virus (GFkV), grapevine fanleaf virus (GFLV) and Arabis mosaic virus (ArMV). Eighty-eight % of the samples were positive for at least one virus, and all five viruses were detected, thought with different incidence, i.e. GLRaV-3 (84%), GFLV (43%), GLRaV-1 (14%), GFk…

Transgene detection by digital droplet PCR

2014

Somatic gene therapy is a promising tool for the treatment of severe diseases. Because of its abuse potential for performance enhancement in sports, the World Anti-Doping Agency (WADA) included the term 'gene doping' in the official list of banned substances and methods in 2004. Several nested PCR or qPCR-based strategies have been proposed that aim at detecting long-term presence of transgene in blood, but these strategies are hampered by technical limitations. We developed a digital droplet PCR (ddPCR) protocol for Insulin-Like Growth Factor 1 (IGF1) detection and demonstrated its applicability monitoring 6 mice injected into skeletal muscle with AAV9-IGF1 elements and 2 controls over a 3…

On the maximum magnetic field amplification by the magnetorotational instability in core-collapse supernovae

2016

Whether the magnetorotational instability (MRI) can amplify initially weak magnetic fields to dynamically relevant strengths in core collapse supernovae is still a matter of active scientific debate. Recent numerical studies have shown that the first phase of MRI growth dominated by channel flows is terminated by parasitic instabilities of the Kelvin-Helmholtz type that disrupt MRI channel flows and quench further magnetic field growth. However, it remains to be prop- erly assessed by what factor the initial magnetic field can be amplified and how it depends on the initial field strength and the amplitude of the perturbations. Different termination criteria leading to different estimates of…

Induction of tcI 7 , a gene encoding a β-subunit of proteasome, in tobacco plants treated with elicitins, salicylic acid or hydrogen peroxide 1

2000

We previously isolated, by differential display and 5′ RACE (rapid amplification of cDNA ends), cDNAs corresponding to genes activated following cryptogein treatment of tobacco cell suspensions, among them tcI 7 (tcI for obacco ryptogein nduced), a gene encoding a β-subunit of proteasome. Here, we report that tcI 7 was up-regulated in tobacco plants treated with elicitins (cryptogein and parasiticein) that have been shown to induce a systemic acquired resistance (SAR). Moreover, subsequent inoculation of tobacco with the pathogen Phytophthora parasitica var. nicotianae (Ppn) was shown to induce an additional activation of tcI 7 in tobacco plants pretreated with cryptogein. We also showed an…

Frequency and prognostic impact of ALK amplifications and mutations in the European Neuroblastoma Study Group (SIOPEN) high-risk neuroblastoma trial …

2021

Purpose: In neuroblastoma (NB), the ALK receptor tyrosine kinase can be constitutively activated through activating point mutations or genomic amplification. We studied ALK genetic alterations in high-risk (HR) patients on the HR-NBL1/SIOPEN trial to determine their frequency, correlation with clinical parameters, and prognostic impact. Materials and methods: Diagnostic tumor samples were available from 1,092 HR-NBL1/SIOPEN patients to determine ALK amplification status (n = 330), ALK mutational profile (n = 191), or both (n = 571). Results: Genomic ALK amplification (ALKa) was detected in 4.5% of cases (41 out of 901), all except one with MYCN amplification (MNA). ALKa was associated with …

Cloud top altitude retrieved from Lidar measurements during ACLOUD at 1 second resolution

2021

During the ACLOUD aircraft campaign (23.5.2017 - 26.6.2017) the AMALi Lidar was installed mostly nadir pointing. This dataset contains the cloud top altitude from those measurements (altitudes with a strong signal increase) as well as a cloud mask, derived from the optical depth of the column at 1 second resolution. The majority of the data was collected northwest of the Svalbard archipelago. More details on the campaign can be found in Wendisch 2018 and Ehrlich 2019 and here (https://home.uni-leipzig.de/~ehrlich/ACLOUD_wiki_doku). Please check the data documentation (https://download.pangaea.de/reference/108729/attachments/readme_documentation_AMALi_cloudtop.pdf) before using this dataset.
