The “mode” of lymphocyte extravasation through HEV of Peyer's patches and its role in normal homing and inflammation
2007
The mode of lymphocyte transendothelial migration in the postcapillary high endothelial venules (HEVs) of Peyer's patches during normal homing and acute inflammation in the guinea pig was studied. It is common opinion that the lymphocyte transendothelial passage from the blood stream into the extravasal lymphoid tissue calls for a multistep process of endothelial and lymphocyte molecules favoring tethering, rolling, activation, arrest and its firm adhesion to the endothelial luminal surface. Ultrastructural serial pictures and the three-dimensional reconstruction of HEVs with lymphocytes during different moments of their transmigration through the endothelial wall enabled us to demonstrate …
Sodium/hydrogen exchange inhibition with cariporide reduces leukocyte adhesion via P-selectin suppression during inflammation
2008
Background and purpose: The Na+/H+ exchange (NHE) inhibitor cariporide is known to ameliorate ischaemia/reperfusion (I/R) injury by reduction of cytosolic Ca2+ overload. Leukocyte activation and infiltration also mediates I/R injury but whether cariporide reduces I/R injury by affecting leukocyte activation is unknown. We studied the effect of cariporide on thrombin and I/R induced leukocyte activation and infiltration models and examined P-selectin expression as a potential mechanism for any identified effects. Experimental approach: An in vivo rat mesenteric microcirculation microscopy model was used with stimulation by thrombin (0.5 μ ml−1) superfusion or ischaemia (by haemorrhagic shock…
Glutathione in Cancer Biology and Therapy
2006
The glutathione (GSH) content of cancer cells is particularly relevant in regulating mutagenic mechanisms, DNA synthesis, growth, and multidrug and radiation resistance. In malignant tumors, as compared with normal tissues, that resistance associates in most cases with higher GSH levels within these cancer cells. Thus, approaches to cancer treatment based on modulation of GSH should control possible growth-associated changes in GSH content and synthesis in these cells. Despite the potential benefits for cancer therapy of a selective GSH-depleting strategy, such a methodology has remained elusive up to now. Metastatic spread, not primary tumor burden, is the leading cause of cancer death. Fo…
Insights in ChAdOx1 nCoV-19 vaccine-induced immune thrombotic thrombocytopenia
2021
Abstract SARS-CoV-2 vaccine ChAdOx1 nCoV-19 (AstraZeneca) causes a thromboembolic complication termed vaccine-induced immune thrombotic thrombocytopenia (VITT). Using biophysical techniques, mouse models, and analysis of VITT patient samples, we identified determinants of this vaccine-induced adverse reaction. Super-resolution microscopy visualized vaccine components forming antigenic complexes with platelet factor 4 (PF4) on platelet surfaces to which anti-PF4 antibodies obtained from VITT patients bound. PF4/vaccine complex formation was charge-driven and increased by addition of DNA. Proteomics identified substantial amounts of virus production-derived T-REx HEK293 proteins in the ethyle…
Oral N-acetylcysteine attenuates the rat pulmonary inflammatory response to antigen.
2003
Oxidative stress is involved in the pathophysiology of inflammatory airway diseases including asthma; therefore, antioxidants might be of clinical benefit in asthma treatment. In the present study, the effects of N-acetylcysteine on sensitised brown Norway rats were examined. N-Acetylcysteine (3 mmol kg body weight(-1) administered orally) was given daily for 1 week before challenge and various antigen-induced pulmonary responses were studied. Antigen exposure increased lipid peroxidation in bronchoalveolar lavage fluid (BALF) and oxidised glutathione levels in lung tissue 2 h after challenge. Lung nuclear transcription factor-KB-binding activity was increased 2 h after challenge, and BALF …

Laparoscopic Treatment of Endoscopic Sigmoid Colon Perforation
2006
Perforation of the colon after colonoscopy is a rare but potentially lethal complication. It usually occurs when endoscopy is performed for therapeutic purposes. In these cases the election of the best treatment is difficult and still controversial. Laparoscopy is a new approach for diagnosis and treatment of this condition. We report a case of a patient who was initially treated laparoscopically after a colonoscopic perforation. A postoperative leak was detected and the patient underwent open surgery. Possible therapeutic approaches and a literature review are discussed.
Stan techniczny i wykorzystanie zabudowań po klasztorach skasowanych w guberni wołyńskiej w latach 1832–1853 w świetle raportu wołyńskiego gubernator…
2019
W rzeczywistości przejęte ziemskie dobra klasztorne zasiliły skarb publiczny, a mienie z różnych powodów niezbywalne powiększyło majątek, pozostający w dyspozycji władz gubernialnych oraz Cerkwi prawosławnej. Władze długo nie doceniały znaczenia zabudowań poklasztornych. Dla większości z nich nie znaleziono nowego użytko…

Attività di impiego e di testing di armi anti-satellite e diritto internazionale
2022
Anti-satellite weapons (ASAT) are one of the major challenges to international security in the «Fourth Domain», namely outer space. This was proved by the test made by Russia in November 2021 and the reaction of the international community. The present testing of said arms against its own satellites, as well as the future and probable operating use of said arms against satellites of other States, raises the problem of their compatibility with international law. In particular, it is being debated in various international fora the legality of ASATs, whose use can generate space debris further congesting terrestrial orbits, thus interfering (and jeopardizing) the activities of other States. Th…
