Novel σ1 antagonists designed for tumor therapy: Structure – activity relationships of aminoethyl substituted cyclohexanes

Abstract Depending on the substitution pattern and stereochemistry, 1,3-dioxanes 1 with an aminoethyl moiety in 4-position represent potent σ1 receptor antagonists. In order to increase the stability, a cyclohexane ring first replaced the acetalic 1, 3-dioxane ring of 1. A large set of aminoethyl substituted cyclohexane derivatives was prepared in a six-step synthesis. All enantiomers and diastereomers were separated by chiral HPLC at the stage of the primary alcohol 7, and their absolute configuration was determined by CD spectroscopy. Given the increased interest in non-pollen palynomorphs (microscopic objects other than pollen identified from pollen slides) in palaeoecological studies, it is necessary to seek a deeper understanding of the reliability of these results. We combined quantitative information of algal pigments and sedimentary ancient DNA (sedaDNA) of phylotaxonomical resolution to validate the richness and abundance of fossil algae in the sediment of a small temperate lake. Manipulating mtDNA in vivo reprograms metabolism via novel response mechanisms.

Mitochondria have been increasingly recognized as a central regulatory nexus for multiple metabolic pathways, in addition to ATP production via oxidative phosphorylation (OXPHOS). Here we show that inducing mitochondrial DNA (mtDNA) stress in Drosophila using a mitochondrially-targeted Type I restriction endonuclease (mtEcoBI) results in unexpected metabolic reprogramming in adult flies, distinct from effects on OXPHOS. Carbohydrate utilization was repressed, with catabolism shifted towards lipid oxidation, accompanied by elevated serine synthesis. Darba mērķis ir papildināt ģenētiskos algoritmus, lai uzlabotu to spēju risināt ceļojošā tirgoņa problēmu. Darba uzdevumi ir izpētīt ģenētisko algoritmu papildināšanas iespējas, izveidot pamata risinājumu, veikt tā papildināšanu un novērtēt tā rezultativitāti izvirzītās problēmas risināšanā.
