Skeletal muscle fibre types, enzyme activities and physical performance in young males and females

Differences in skeletal muscle characteristics, metabolic profiles and functional performance between males and females were investigated using young (15--24 yrs) male and female twins as subjects. The comparison included such variables as anthropometry, muscle strength, mechanical power, maximum oxygen uptake, electrical activation of muscle, muscle fibre composition (m. vastus lateralis), and activities of several skeletal muscle enzymes. Hypertensive obese subjects with glucose intolerance have hyperinsulinaemia, insulin resistance and intracellular cation imbalance resulting in increased sodium content. The aim of our study was to assess in these patients plasma levels of endogenous digoxin-like factor (EDLF), an inhibitor of the sodium-pump mechanism. Oxidative stress has been related to the development of diabetic neuropathy. Experimental diabetes (alloxan injection to mice) promotes early biochemical changes in peripheral nervous tissue, e.g., decrease in Na,K-ATPase activity and glutathione (GSH) peroxidase (GSHPx) activity. The former decrease can be reverted by inhibiting protein kinase C (PKC), since it has been reported that PKC is activated in these experimental conditions. The effect of alloxan-induced diabetes on glutathione peroxidase (GSH-Px) activity in sciatic nerve of mice has been studied. We have found, 7 days after alloxan treatment, a significant decrease in this enzymatic activity in the cytosol of sciatic nerve of diabetic mice, and moreover, that these changes remained unaltered up to 21 days after alloxan injection. No modification in the glutathione content of sciatic nerve of diabetic mice was observed throughout the experiment when compared with controls. Abstract Plasmalemma vesicles isolated from rice (Oryza sativa L.) shoots and roots were used to study the effect of different nickel and cadmium concentrations on the Mg2+-ATPase activity. In vivo application of the metals for 5 or 10 days resulted in a stimulation of the enzyme activity in shoots, while either increases or decreases were observed in roots depending on the concentration and the type of ion tested. In contrast, an inhibitory effect was always observed on shoot and root plasmalemma ATPase activity when these ions were applied in vitro. Many biomolecules are esterified with carbonic acid, phosphoric acid or sulphuric acid. The enzymes responsible for the hydrolytic cleavage of the esters are the carboxylester hydrolases, phosphoesterases and sulphatases. In addition to the phosphomonoesterases, we will also consider the phosphodiesterases and nucleases that are specific for cAMP and cGMP, as well as the ATPases that cleave phosphoric acid anhydride bonds. The ion-transporting ATPases will be discussed together with comparative biochemical data on ion channels. The chapter ends with a discussion of carboanhydrases.

The archaebacterium Thermoplasma acidophilum is cultivated at 59 degrees C in a medium containing sulfuric acid of pH 2. The purified bipolar membrane spanning main phospholipid (MPL) of this organism can be used to produce stable liposomes of 100-500 nm in diameter either using a French pressure cell detergent dialysis or sonication.
