Magnetic resonance imaging of viral particle biodistribution in vivo

2006

We describe here a technique for the visualization of viral vector delivery by magnetic resonance imaging (MRI) in vivo. By conjugating avidin-coated baculoviral vectors (Baavi) with biotinylated ultra-small superparamagnetic iron oxide particles (USPIO), we are able to produce vector-related MRI contrast in the choroid plexus cells of rat brain in vivo over a period of 14 days. Ten microlitres of 2.5 x 10(10) PFU/ml nuclear-targeted LacZ-encoding Baavi with bUSPIO coating was injected into rat brain ventricles and visualized by MRI at 4.7 T. As baculoviruses exhibit restricted cell-type specificity in the rat brain, altered MRI contrast was detected in the choroid plexus of the injected ve…

Modifying the body distribution of HPMA-based copolymers by molecular weight and aggregate formation.

2011

There is a recognized need to create well-defined polymer probes for in vivo and clinical positron emission tomography (PET) imaging to guide the development of new generation polymer therapeutics. Using the RAFT polymerization technique in combination with the reactive ester approach, here we have synthesized well-defined and narrowly distributed N-(2-hydroxypropyl)methacrylamide homopolymers (pHPMA) (P1* and P2*) and random HPMA copolymers consisting of hydrophilic HPMA and hydrophobic lauryl methacrylate comonomers (P3* and P4*). The polymers had molecular weights below (P1* and P3*) and above the renal threshold (P2* and P4*). Whereas the homopolymers dissolve in isotonic solution as in…

Genetic relationships between sympatric and allopatric Coregonus ciscoes in North and Central Europe

2021

Abstract Background Sympatric speciation along ecological gradients has been studied repeatedly, in particular in freshwater fishes. Rapid post-glacial ecological divergence has resulted in numerous endemic species or ecologically distinct populations in lakes of the temperate zones. Here, we focus on the Baltic cisco (Coregonus albula) complex, to study the genetic similarity among two pairs of sympatric autumn- and spring-spawning populations from post-glacial German Lakes Stechlin and Breiter Luzin. For comparison, we included a similar pair of sympatric populations from the Swedish Lake Fegen. We wanted to explore potential genetic similarities between the three sympatric cisco populati…

Intake and milk production of goats grazing Sulla forage under different stocking rates

2010

This investigation aimed to individuate the dietary factors affecting the milk urea nitrogen (MUN) concentration in goats grazing herbaceous pasture and, particularly, to verify the relationship linking the diet crude protein (CP) content to MUN. A total of 205 individual observations regarding dietary and milk variables of 37 Girgentana goats involved in two experiments were used. Goats, averaging 154±14 days in milk and 38.1±5.4 kg of live weight, grazed on swards and received 500 g/d of barley meal. Sward biomass, herbage selected by goats and individual milk yield were measured and sampled weekly. The herbage intake and diet digestibility were estimated by the n-alkane method. Milk urea…

Low-fouling, mixed-charge poly-L-lysine polymers with anionic oligopeptide side-chains

2018

Biosensors and biomedical devices require antifouling surfaces to prevent the non-specific adhesion of proteins or cells, for example, when aiming to detect circulating cancer biomarkers in complex natural media (e.g., in blood plasma or serum). A mixed-charge polymer was prepared by the coupling of a cationic polyelectrolyte and an anionic oligopeptide through a modified "grafting-to" method. The poly-l-lysine (PLL) backbone was modified with different percentages (y%) of maleimide-NHS ester chains (PLL-mal(y%), from 13% to 26%), to produce cationic polymers with specific grafting densities, obtaining a mixed-charge polymer. The anionic oligopeptide structure (CEEEEE) included one cysteine…
