TMOD-36. PRECISE INVESTIGATION OF CANCER STEM CELLS IN A MOUSE GLIOBLASTOMA MODEL
2018
Cancer stem cells (CSCs) have been shown to play a critical role in glioblastoma (GBM) pathogenesis. However, a precise and thorough understanding of these cells is still lacking. Here we design a novel mouse model to label, purify, and study cancer stem cells in vivo. Firstly we generate and characterize a new transgene to label neural stem/progenitor cells in the subventricular zone (SVZ) with GFP, and drive expression of CreERT2 and human diphtheria toxin receptor in the same cells (CGD: nestin-CreERT2-H2BeGFP-hDTR). Following analysis with both bulk and single cell RNA sequencing of the SVZ tissue demonstrate its faithful expression in the stem/progenitor cell compartment. We then cross…
TMIC-49. POTASSIUM CHANNEL KIR4.1 AND GLUTAMINE SYNTHETASE ARE DYSREGULATED IN GLIOMA
2017
The potassium channel KIR4.1 (KCNJ10) and the glutamate catalyzing enzyme glutamine synthetase (GS) are highly expressed in glial cells of the central nervous system. Both glial proteins play important roles in the maintenance of neuronal activity and neurotransmission. Dysfunction of both proteins can result in altered neuronal excitability and may lead to excitotoxicity. We analyzed 35 snap frozen tissue blocks (glioblastoma [GBM], n=22; low grade astrocytoma (LGA), n=8; oligodendroglioma (OG), n=3; oligoastrocytoma, n=2). All glioma samples had a matching tissue specimen from both the tumor core and the adjacent normal-appearing infiltration zone. Molecular subtyping (MGMT, IDH1/2, 1p/19…
Gene Set to Diseases (GS2D): disease enrichment analysis on human gene sets with literature data
2016
Large sets of candidate genes derived from high-throughput biological experiments can be characterized by functional enrichment analysis. The analysis consists of comparing the functions of one gene set against that of a background gene set. Then, functions related to a significant number of genes in the gene set are expected to be relevant. Web tools offering disease enrichment analysis on gene sets are often based on gene-disease associations from manually curated or experimental data that is accurate but does not cover all diseases discussed in the literature. Using associations automatically derived from literature data could be a cost effective method to improve the coverage of disease…
Evaluation of nutritional parameters in infant formulas and powdered milk by Raman spectroscopy
2007
It has been made a critical evaluation of the application of near infrared Fourier transform-Raman spectroscopy for the simultaneous determination of the most important nutritional parameters such as energetic value, carbohydrate, protein and fat contents of infant formula and powdered milk samples based on the use of partial least squares (PLS) regression analysis. A highly heterogeneous population of 23 samples, covering a wide range of infant food formula and powdered milk, were obtained from the Spanish market. Raman spectra, obtained by excitation with a Nd:YAG laser at 1064 nm, show no disturbing fluorescence effects; therefore sample spectra can be recorded without any previous prepa…
Inverse estimation of terminal connections in the cardiac conduction system
2017
Modeling the cardiac conduction system is a challenging problem in the context of computational cardiac electrophysiology. Its ventricular section, the Purkinje system, is responsible for triggering tissue electrical activation at discrete terminal locations, which subsequently spreads throughout the ventricles. In this paper, we present an algorithm that is capable of estimating the location of the Purkinje system triggering points from a set of random measurements on tissue. We present the properties and the performance of the algorithm under controlled synthetic scenarios. Results show that the method is capable of locating most of the triggering points in scenarios with a fair ratio bet…
Role of a medication in polyurethane foam in the treatment of diabetic foot lesions
2010
Background The treatment of diabetic foot lesions need medications capable of managing wound infection and exudates that, variously combined, can influence the natural history of the pathology and therefore limb survival. A particular and economic device, is represented by honeycomb structure polyurethane foam Ligasano ©.
Cartoon filter via adaptive abstraction
2016
We propose a non-parametric methodology to realize abstraction images. The redundant wavelet "a trous" algorithm is applied for details detection. An multi-scale circular median filter is used as a smoothing filter. The proposed algorithm is simple and fast on low-cost entry-level hardware. Abstraction in computer graphics defines a procedure that discriminates the essential information that is worth keeping. Usually details, that correspond to higher frequency components, allow to distinguish otherwise similar images. Vice versa, low frequencies are related to the main information, which are larger structures. Contours themselves may also be identified by high frequencies and separate each pi…
A novel method for harmonic mitigation for single-phase five-level cascaded H-Bridge inverter
2018
The efficiency of a system is a very important parameter for high power electrical drives applications. Moreover, the efficiency of power converters plays a fundamental role. Aim of this work, is to propose a novel selective harmonic mitigation method without solving non-linear equations. Through a very simple approach, the polynomial equations which drive the control angles have been detected for a single-phase five-level cascaded H-Bridge inverter. The obtained polynomial equations can be easily implemented in a digital system to real-time operation. The paper also presents the simulation analysis and experimental validation.
Analysis of neuronal networks in the visual system of the cat using statistical signals--simple and complex cells. Part II.
1978
Superimposing additively a two-dimensional noise process to deterministic input signals (bars) the neurons of area 17 show a class-specific reaction for the task of signal extraction. Moving both parts of the signals simultaneously and varying the signal to noise ratio (S/N) the simple cells achieve the same performance as resulted from the psychophysical experiment. Type I complex cells extract moving deterministic signals (i.e. bars) from the stationary noise, whereas in the answers of Type II complex cells the statistical parts of the signals predominate. Considering the different cell types each as a series of a linear and a nonlinear system one obtains the cell specific space-time freq…
Analysis of neuronal networks in the visual system of the cat using statistical signals
1976
If the input signals of the visual system in the cat are statistical patterns in space and time, a complete system analysis can be carried out. What counts here as a system are the neuronal networks between retina and recording site. In the case of linearity, one obtains the temporal impulse response functions at every point in the receptive field with the aid of correlation methods. The measuring time is about one minute. Some aspects of the procedure are explained in terms of examples. The method of measurement also makes it possible to determine the characteristic function of the system in time and space between different recording sites within the cortex. It is possible to specialize th…
