Enhancement of hepatitis C virus RNA replication by cell culture-adaptive mutations.

ABSTRACT Studies of the Hepatitis C virus (HCV) replication cycle have been made possible with the development of subgenomic selectable RNAs that replicate autonomously in cultured cells. In these replicons the region encoding the HCV structural proteins was replaced by the neomycin phosphotransferase gene, allowing the selection of transfected cells that support high-level replication of these RNAs. Subsequent analyses revealed that, within selected cells, HCV RNAs had acquired adaptive mutations that increased the efficiency of colony formation by an unknown mechanism. This paper takes into consideration Timberlake Wertenbaker's adaptation of Ferrante's My Brilliant Friend (2016) for the radio. In particular, the focus of the paper is on the first of the four radioplays and on the dramatic strategies used and adaptation choices made by Wertenbaker. We shall focus on the use of two main adult voices (Lena and Lila), the use of the voice over, and the representation of time. The chapter also considers the image of "neapolitanity" offered in the radioplay, the reception with the British audience with respect to the meridionist idea of Southern Italy as poor and violent. Purpose: The authors investigated the effects of sex, energy availability (EA), and health status on the change in hemoglobin mass (ΔHbmass) in elite endurance athletes over ∼3–4 wk of live-high–train-high altitude training in Flagstaff, AZ (2135 m; n = 27 women; n = 21 men; 27% 2016 Olympians). Modeling multipartite virus evolution: the genome formula facilitates rapid adaptation to heterogeneous environments

Multipartite viruses have two or more genome segments, and package different segments into different particle types. Although multipartition is thought to have a cost for virus transmission, its benefits are not clear. Recent experimental work has shown that the equilibrium frequency of viral genome segments, the setpoint genome formula (SGF), can be unbalanced and host-species dependent. These observations have reinvigorated the hypothesis that changes in genome-segment frequencies can lead to changes in virus-gene expression that might be adaptive.
