SEQUENTIALLY ADJUSTED RANDOMIZATION TO FORCE BALANCE IN CONTROLLED TRIALS WITH UNKNOWN PREVALENCE OF COVARIATES: APPLICATION TO ALCOHOLISM RESEARCH
2005
In treatment outcome studies with small to medium sample sizes (n200), the balance of groups with regard to important factors, which sometimes occur at low prevalence, is indispensable for adequate interpretation. This study tested a method for use in clinical alcoholism research, an uncomplicated procedure for satisfactory randomization of patients to different treatments, taking into account relevant background variables. An easily applicable modification of Efron's biased coin method for the randomization of treatments within strata of unknown but low prevalence was compared with the original approach and alternative methods by computer simulation (10,000 runs). An application example for…
Targeting Homer genes using adeno-associated viral vector: lessons learned from behavioural and neurochemical studies.
2008
Over a decade of in-vitro data support a critical role for members of the Homer family of postsynaptic scaffolding proteins in regulating the functional architecture of glutamate synapses. Earlier studies of Homer knockout mice indicated a necessary role for Homer gene products in normal mesocorticolimbic glutamate transmission and behaviours associated therewith. The advent of adeno-associated viral vectors carrying cDNA for, or short hairpin RNA against, specific Homer isoforms enabled the site-directed targeting of Homers to neurons in the brain. This approach has allowed our groups to address developmental issues associated with conventional knockout mice, to confirm active roles for di…
2015
Treatment with acamprosate is a valid tool to complement psychotherapy as it does not cause addiction, abuse or withdrawal of its suspension and does not interfere with other medications that patients often alcoholics must take. To evaluate the effectiveness, our study evaluated the effects of Acamprosate compared to γ-hydroxybutyrate in clinical-physiological and social health in a way indicator of a possible therapeutic success in terms of abstinence from alcohol and social reintegration. A total of 36 patients were observed, of which 5, 4 men and 1 woman at the Drug Addiction Service (Servizio Tossicodipendenze, Ser.T) Alcamo, and 31, 21 men and 10 women at the Ser.T of Palermo. As regar…
Biomarkers to disclose recent intake of alcohol: potential of 5-hydroxytryptophol glucuronide testing using new direct UPLC-tandem MS and ELISA metho…
2007
Aims: This study compared two new methods for direct determination of 5-hydroxytryptophol glucuronide (GTOL) in urine, a biomarker for detection of recent alcohol consumption. Methods: Urine samples were collected from ten alcoholic patients during recovery from intoxication. A direct injection ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method for measurement of the urinary GTOL to 5-hydroxyindoleacetic acid (5-HIAA) ratio, and an ELISA assay for direct measurement of GTOL, were used. Comparison was made with the urinary ethanol and ethyl glucuronide (EtG) concentrations. Results: The breath ethanol concentration on admission ranged between 1.0-3.1 g/l. Th…
Predictors of weekly alcohol drinking and alcoholrelated problems in binge-drinking undergraduates
2015
Testing Three Theories of Cognitive Dysfunction in Problem Alcohol Use
2014
- Objectives : many hypotheses concerning the cognitive functions affected by chronic alcoholism have been advanced. The aim of this study was to test cognitive performance in a sample of alcoholics and to examine results comparing three different models: the frontal lobe hypothesis, the lateralization hypothesis and the diffuse brain hypothesis. - Methods : 51 patients who refer to alcoholism service have been included in this study. Cognitive functions were assessed by trained psychologist with a specific neuropsychological battery. -Results : our results show a ranking of impairment degrees in different cognitive functions and different influence of clinical features. In our sample the f…

Cognitive deficits related to alcohol abuse: a preliminary study
2012
-Objective : cognitive deficits may compromise patients’ utilization of rehabilitative information. There is an increasing attention on the opportunity to integrate a specific support to cognitive functions in alcohol detox programme and the first step to program the clinical intervention is to have a complete overview of cognitive deficits in alcoholics clinical sample. -Methods : 62 patients who refer to alcoholism service were screened at admission time. Cognitive functions were assessed by trained psychologist with a specific neuropsychological battery. The impact of clinical features as age, gender, scholarship and concurrent polyabuse was evaluated for each function. -Results : the 45…
