New derivatives of progesterone and aldosterone were synthesized and functionally tested with commercially available antibodies. The covalent labelling of antibodies specific for aldosterone and progesterone was detected by SDS/PAGE analysis and subsequent autoradiography after using 3-(O-carboxymethyl)-oximino-(3-[125I]iodo-4-azidosalicylamidobu tylamine) derivatives of aldosterone and progesterone, respectively, as photoactivatable radioligands. Primary aldosteronism (PA) is the most common endocrine form of hypertension and may carry an increased risk of atrial flutter or fibrillation (AFF). The primary goal of this multicentre cohort study is thus to prospectively establish the prevalence of PA in consecutive hypertensive patients referred for lone (non-valvular), paroxysmal or permanent AFF. Summary Groups of male rats were treated with antihypertensive drugs (alpha-methyl-dopa, clonidine, propranolol, reserpine, diazoxide) which were administered under conditions causing the onset of high plasma renin activity (PRA) and high plasma and urine aldosterone levels, i.e. together with distilled water load (5% of body weight). Alpha-methyl-dopa and propranolol, which fail to significantly alter PRA and plasma aldosterone levels when administered without distilled water load, cause a marked decrease of plasma and urine aldosterone levels and of PRA when administered together with distilled water load, while diazoxide, and reserpine cause a marked increase of these values. Summary The data reported in the present paper refer to quantitative variations of plasma renin activity and plasma aldosterone levels after administration of alpha-blocking agents, i.e. phentolamine, phenoxybenzamine and gihydroergotoxin derivatives, either to rats kept on a standard diet with water ad libitum or to rats receiving distilled water load, the latter treatment causing an increase of both plasma renin activity and plasma aldosterone levels if compared to control values. Chromatographic and mass spectrometric characteristics of 20-dihydroaldosterone.

The 20 alpha-reduced derivative of aldosterone, 20 alpha-dihydroaldosterone, was needed as reference compound in order to continue the studies on 18-hydroxylation in the Y-1 adrenal cell line. It was obtained by reduction of aldosterone with sodium borohydride. Analysis of the products of the reaction as methoxime trimethylsilyl (MO-TMS) derivatives by gas chromatography (GC) and GC-mass spectrometry (GC-MS) showed three possible forms of the compound. Their identification was confirmed by comparison with the products obtained by stereospecific reduction of aldosterone using 3 alpha,20 beta-hydroxysteroid dehydrogenase.
