Myelin changes in Alexander disease

Introduction: Alexander disease (AxD) is a type of leukodystrophy. Its pathological basis, along with myelin loss, is the appearance of Rosenthal bodies, which are cytoplasmic inclusions in astrocytes. Mutations in the gene coding for glial fibrillary acidic protein (GFAP) have been identified as a genetic basis for AxD. However, the mechanism by which these variants produce the disease is not understood. Development: The most widespread hypothesis is that AxD develops when a gain-of-function mutation causes an increase in GFAP. However, this mechanism does not explain myelin loss, given that experimental models in which GFAP expression is normal or mutated do not exhibit myelin disorders. …

Background Alexander disease (AxD) is a rare disease caused by mutations in the gene encoding glial fibrillary acidic protein (GFAP). The disease is characterized by presence of GFAP aggregates in the cytoplasm of astrocytes and loss of myelin. Objectives Determine the effect of AxD-related mutations on adult neurogenesis. Methods We transfected different types of mutant GFAP into neurospheres using the nucleofection technique. Results We find that mutations may cause coexpression of GFAP and NG2 in neurosphere cultures, which would inhibit the differentiation of precursors into oligodendrocytes and thus explain the myelin loss occurring in the disease. On orderability of fibred knot groups

It is known that knot groups are right-orderable, and that many of them are not bi-orderable. Here we show that certain bred knots in S 3 (or in a homology sphere) do have bi-orderable fundamental group. In particular, this holds for bred knots, such as 41, for which the Alexander polynomial has all roots real and positive. This is an application of the construction of orderings of groups, which are moreover invariant with respect to a certain automorphism.

We show that the image of a proper closed continuous map, f , from an n -manifold X to an ( n + 1)-manifold Y , such that H 1 (Y; Z 2 ) =0 , separates Y into at least two connected components provided the self-intersections set of f is not dense in any connected component of Y . We also obtain some geometrical applications.

A partir de fuentes inéditas, este artículo analiza qué tipo de medidas administrativas estableció la Corona en la década de 1580 para someter la gestión financiera de la capitanía general del ejército de Flandes a un control más riguroso. también proporciona evidencias documentales de por qué la Corona desistió en su empeño fiscalizador en esta misma década.
