Observation of the Early Structural Changes Leading to the Formation of Protein Superstructures.
2014
Formation of superstructures in protein aggregation processes has been indicated as a general pathway for several proteins, possibly playing a role in human pathologies. There is a severe lack of knowledge on the origin of such species in terms of both mechanisms of formation and structural features. We use equine lysozyme as a model protein, and by combining spectroscopic techniques and microscopy with X-ray fiber diffraction and ab initio modeling of Small Angle X-ray Scattering data, we isolate the partially unfolded state from which one of these superstructures (i.e., particulate) originates. We reveal the low-resolution structure of the unfolded state and its mechanism of formation, hi…
Real-world experience with obeticholic acid in patients with primary biliary cholangitis
2021
Background & aims Obeticholic acid (OCA) is the second-line treatment approved for patients with primary biliary cholangitis (PBC) and an inadequate response or intolerance to ursodeoxycholic acid. We aimed to evaluate the effectiveness and safety of OCA under real-world conditions. Methods Patients were recruited into the Italian PBC Registry, a multicentre, observational cohort study that monitors patients with PBC at national level. The primary endpoint was the biochemical response according to Poise criteria; the secondary endpoint was the biochemical response according to normal range criteria, defined as normal levels of bilirubin, alkaline phosphatase (ALP), and alanine aminotransfer…
Rapid and sensitive detection of metapneumovirus in clinical specimens by indirect fluorescence assay using a monoclonal antibody.
2008
Human metapneumovirus, with two known genotypes named A and B, is associated with mild respiratory symptoms to severe LRTI in children, high-risk adults and the elderly. Rapid and reliable methods of hMPV detection in clinical samples are essential to implement appropriate care, to better understand the pathology of hMPV and to determine its epidemiology. Respiratory samples from 1,386 patients collected during 2 consecutive years were screened for hMPV using indirect immunofluorescence (IFA) assay with a monoclonal antibody. Forty-three patients tested positive for hMPV by the IFA method. In parallel, the samples were examined with RT-PCR on the F gene. Of these, 41 specimens were RT-PCR p…
Anatomical and electrophysiological study of the vomeronasal circuits: Amygdaloid response to odours and pheromones
2017
Rodents detect information concerning the world around them mainly through two chemosensory systems: the olfactory and the vomeronasal systems. In order to develop an appropriate behavioural response to their environment, these systems exhibit both functional and physiological convergence. Further understanding of the organization and function of the olfactory systems would allow us to comprehend how their information is integrated in the brain. In a first approach we performed a thorough analysis of the connections of key structures involved in the processing of vomeronasal information: the medial (Me) and the posteromedial cortical (PMCo) amygdaloid nucleus. Then, we enquire the populatio…
