In vitro fertilization and andrology laboratories in 2030

The in vitro fertilization and andrology laboratories are at the center of assisted reproductive technologies and the place where technicians and embryologists manipulate gametes and preimplantation-stage embryos with the goal of achieving the best embryo for transfer. Through the years, these laboratories have seen developments in technique, technology, and testing. The goal of this Views and Interviews series is to bring together the thought leaders in the field and envision what the laboratories will look like in the next 10 years.

Fertilization and early embryology: Morphometric characterization of normal and abnormal human zygotes

Human zygotes (n = 278) from 96 in-vitro fertilization gonadotrophin-stimulated cycles were photographed in their pronuclear stage (16-18 h post-insemination). Normal morphological fertilization (two pronuclei) was observed in 215 zygotes, 17 showed only one pronucleus, 40 showed three pronuclei and six showed four. Area, perimeter and maximum and minimum diameters of each zygote and pronucleus were measured using an IBAS 2000 (Kontron) image analyser. When the four groups were compared, whole zygotes did not show any morphometric difference. A new insect ovicide (Ov. 165049), 4-methoxy-benzoic acid propargylester, synthesized by BASF (Ludwigshafen) was tested on Oncopeltus fasciatus and Manduca sexta. In the case of O. fasciatus also the effects on last-instar larvae and adult bugs were studied. Topical application of 40 μg/larva dissolved in acetone caused a mortality exceeding 50% at day 1 after treatment. The few animals which developed into imagines did not reproduce. A treatment with 4 μg/larva had no severe impact on metamorphosis and reproduction. Adult O. fasciatus received doses of 40 μg, 20 μg and 4 μg per animal. Summary answer No correlation between CMI in live motile spermatozoa, DNA Fragmentation Index (DFI) and other conventional seminal parameters were found in male infertile patients.
