A morphometric tool applied to angiogenesis research based on vessel segmentation

2013

Given that angiogenesis and lymphangiogenesis are strongly related to prognosis in neoplastic and other pathologies, and that some existing methods for their study provide different results, we aim to construct a morphometric tool to allow complete and accurate quantification and measurement of different aspects of the shape and size of vascular vessels.

Perspectives in Clinical Hemorheology and Microcirculation: Review of the Conference of the German Society for Clinical Microcirculation and Hemorheo…

2011

During the 29th conference of the German Society for Clinical Microcirculation and Hemorheology – held in the Freie Universitat Berlin a broad range of different topics of experimental and clinical studies in the field were presented. A special spotlight was dedicated to the Conference President’s (Prof. Dr. J. Plendl, Head of the Department of Veterinary Anatomy) scientific area “Anatomical and Experimental Microcirculation” [5, 6, 9, 13]. Of special interest were the angiogenetic studies: Examinations of the influence of PDGF-BB on adipose-tissue derived stem cells and how these influence angiogenesis [9] and the angiogenesis and healing with non-shrinking, fast degradable PLGA/CaP scaffo…

Comparative morphometric evaluation of microvessel density and nuclear area in ductal carcinoma in situ and hyperplastic ductal breast lesions

1999

Abstract Tumour angiogenesis, as assayed by microvessel density, has been proposed as a prognostic factor in invasive breast cancer. Angiogenesis in noninvasive tumours and hyperplastic mammary lesion has not been well-established. The aim of the present study was to evaluate the microvessel density in ductal proliferative breast lesions and to investigate the relationships between microvessel density and histopathologic features. In addition, morphometric measurement of nuclear area in ductal proliferative breast lesions and its relationship with microvessel density has been examined. Thirty-four cases were evaluated, 19 ductal carcinomas in situ (DCIS) and 15 of usual and atypical ductal …

Evaluation of the potential therapeutic effects of a double-stranded RNA mimic complexed with polycations in an experimental mouse model of endometri…

2015

Objective To assess the therapeutic potential of polyinosine-polycytidylic acid, a double-stranded RNA molecule with selective proapoptotic and antiangiogenic activity, complexed with polyethyleneimine (pIC PEI ) in treating endometriosis. Design A heterologous mouse model of endometriosis was created by injecting human endometrial fragments into the peritoneum. Endometrial fragments were engineered to express the fluorescent protein mCherry as a reporter to monitor status over the course of the 4-week study. Setting University-affiliated infertility center. Animal(s) Ovariectomized and hormone-replaced nude mice (n = 30) injected with fluorescent-labeled human endometrial fragments at 4–6 …

C1q as a novel player in angiogenesis with therapeutic implication in wound healing

2014

We have previously shown that C1q is expressed on endothelial cells (ECs) of newly formed decidual tissue. Here we demonstrate that C1q is deposited in wound-healing skin in the absence of C4 and C3 and that C1q mRNA is locally expressed as revealed by real-time PCR and in situ hybridization. C1q was found to induce permeability of the EC monolayer, to stimulate EC proliferation and migration, and to promote tube formation and sprouting of new vessels in a rat aortic ring assay. Using a murine model of wound healing we observed that vessel formation was defective in C1qa(-/-) mice and was restored to normal after local application of C1q. The mean vessel density of wound-healing tissue and …

Norrie gene product is necessary for regression of hyaloid vessels.

2004

To investigate the nature and origin of the vitreous membranes in mice with knock-out of the Norrie gene product (ND mice).Eighty-two eyes of ND mice of different age groups (postnatal day [P]0-13 months) and 95 age-matched wild-type control mice were investigated. In vitreoretinal wholemounts and in sagittal sections, vessels and free cells were visualized by labeling for lectin. In addition, staining with a marker for macrophages (F4/80) and collagen XVIII/endostatin known to be involved in regression of hyaloid vessels was performed for light and electron microscopic investigations. Endostatin expression was confirmed by Western blot analysis.Wild-type controls showed the typical pattern…

Angiogenesis of cancer of the cervix. Contrast-enhanced dynamic MRT, histological quantification of capillary density and lymph system infiltration

1998

Purpose: It was the aim of this project to examine (i) the relationships between contrast-enhanced dynamic MR imaging derived characteristics and histologic microvessel density counts – a recognized surrogate of tumor angiogenesis – from tumors in patients with primary or recurrent cancer of the uterine cervix, and (ii) to correlate these parameters with lymphatic involvement (i. e. lymphatic channels) to assess tumorbiological aggressiveness in terms of lymphatic spread. Materials and methods: Pharmacokinetic MR imaging parameters (amplitude A, exchange rate constant k 21) were derived from contrast-enhanced dynamic MR imaging in thirty-three patients with biopsy proven cancer of the uteri…
