Identification of Synaptic Integration Mode in CA3 Pyramidal Neuron Model

2019

International audience; A morphologically realistic and anisotropic model of CA3 pyramidal neuron was developed to determine the synaptic integration modes the neuron is able to perform. Linearity and nonlinearity were identified in different synaptic locations with varying active mechanisms such as the presence of ionic channels in the dendritic arbor and the types of receptors in the synapse. Quantification of synaptic integration was performed using paired-pulse stimulation protocol and subthreshold input/output (sI/O) transformation. Results show that the mode of synaptic integration is location-dependent while the linearity or nonlinearity in the integration is mainly influenced by the…

Putative excitatory and putative inhibitory inputs are localised in different dendritic domains in aDrosophilaflight motoneuron

2012

Input-output computations of individual neurons may be affected by the three-dimensional structure of their dendrites and by the targeting of input synapses to specific parts of their dendrites. However, only few examples exist where dendritic architecture can be related to behaviorally relevant computations of a neuron. By combining genetic, immunohistochemical, and confocal laser scanning methods this study estimates the location of the spike initiating zone and the dendritic distribution patterns of putative synaptic inputs on an individually identified Drosophila flight motorneuron, MN5. MN5 is a monopolar neuron with more than 4000 dendritic branches. The site of spike initiation was e…

Progression is independent of relapse activity in early multiple sclerosis: a real-life cohort study

2022

Portaccio et al. report that in early relapsing-onset multiple sclerosis, progression independent of relapse activity is an important contributor to disability accumulation. Insidious progression occurs even in the earliest disease phases, suggesting that inflammation and degeneration may represent a single disease continuum.Disability accrual in multiple sclerosis may occur as relapse-associated worsening or progression independent of relapse activity. The role of progression independent of relapse activity in early multiple sclerosis is yet to be established. The objective of this multicentre, observational, retrospective cohort study was to investigate the contribution of relapse-associa…

The genetic contribution of the NO system at the glutamatergic post-synapse to schizophrenia: Further evidence and meta-analysis

2013

NO is a pleiotropic signaling molecule and has an important role in cognition and emotion. In the brain, NO is produced by neuronal nitric oxide synthase (NOS-I, encoded by NOS1) coupled to the NMDA receptor via PDZ interactions; this protein-protein interaction is disrupted upon binding of NOS1 adapter protein (encoded by NOS1AP) to NOS-I. As both NOS1 and NOS1AP were associated with schizophrenia, we here investigated these genes in greater detail by genotyping new samples and conducting a meta-analysis of our own and published data. In doing so, we confirmed association of both genes with schizophrenia and found evidence for their interaction in increasing risk towards disease. Our stron…

Mass measurements towards doubly magic Ni-78 : Hydrodynamics versus nuclear mass contribution in core-collapse supernovae

2022

International audience; We report the first high-precision mass measurements of the neutron-rich nuclei 74,75Ni and the clearly identified ground state of 76Cu, along with a more precise mass-excess value of 78Cu, performed with the double Penning trap JYFLTRAP at the Ion Guide Isotope Separator On-Line (IGISOL) facility. These new results lead to a quantitative estimation of the quenching for the N=50 neutron shell gap. The impact of this shell quenching on core-collapse supernova dynamics is specifically tested using a dedicated statistical equilibrium approach that allows a variation of the mass model independent of the other microphysical inputs. We conclude that the impact of nuclear m…

Alle kouluikäisten lasten varhaisten matemaattisten taitojen tukeminen Nallematikka-ohjelmalla

2015

Tämän opinnäytetyön tarkoituksena oli selvittää Nallematikka-intervention soveltuvuutta niiden 5-vuotiaiden lasten tukemiseksi, joilla arveltiin olevan heikot matemaattiset taidot, riski matematiikan oppimisvaikeuteen ja tarve saada varhaisiin matemaattisiin taitoihinsa tukea. Nallematikalla viitataan lasten varhaisten matemaattisten taitojen ryhmämuotoiseen opetusohjelmaan. Tutkimus toteutettiin viidessä pirkanmaalaisessa päiväkotiryhmässä syksyn 2013–kevään 2014 välisenä aikana. Koeryhmään valikoitui 18 ja verrokkiryhmään 6 lasta. Seulontamenetelmänä lasten valinnassa käytettiin Nallematikka-ohjelman ”Lapsen oppimisvalmiuksien kartoitus”- tehtäviä. Varhaisia matemaattisia taitoja mitattii…

A new culturing strategy optimises Drosophila primary cell cultures for structural and functional analyses

2004

Abstract Neurons in primary cell cultures provide important experimental possibilities complementing or substituting those in the nervous system. However, Drosophila primary cell cultures have unfortunate limitations: they lack either a range of naturally occurring cell types, or of mature physiological properties. Here, we demonstrate a strategy which supports both aspects integrated in one culture: Initial culturing in conventional serum-supplemented Schneider's medium (SM 20K ) guarantees acquisition of all properties known from 30 years of work on cell type-specific differentiation in this medium. Through subsequent shift to newly developed active Schneider's medium (SM active ), neuron…
