Detoxication of carcinogenic fjord-region diol epoxides of polycyclic aromatic hydrocarbons by glutathione transferase P1-1 variants and glutathione.
1998
AbstractEpidemiological studies suggest that individuals differing in the expression of allelic variants of the human glutathione transferase (GST) Pi gene differ in susceptibility to chemical carcinogens such as polycyclic aromatic hydrocarbons (PAH). This study reports the catalytic efficiencies (kcat/Km) of two naturally occurring variants, GSTP1-1/I-105 and GSTP1-1/V-105, towards a series of fjord-region diol epoxides representing potent biologically active PAH metabolites, and two GSTP1-1 mutants with Ala105 and Trp105 in the active site. The results indicate that individuals who are homozygous for the allele encoding GSTP1-1/V-105 might be more susceptible to PAH carcinogenesis due to…
Polycyclic aromatic hydrocarbons (PAHs) and their oxygen-containing derivatives (OPAHs) in soils from the Angren industrial area, Uzbekistan.
2009
We measured the concentrations and depth distribution (0-10, 10-20 cm) of 31 PAHs and 12 OPAHs in soils at eleven equidistant sampling points along a 20-km transect in the Angren industrial region (coal mine, power plant, rubber factory, gold mine), Uzbekistan to gain an insight into their concentrations, sources, and fate. Concentrations of all compounds were mostly much higher in the 0-10 cm than in the 10-20 cm layer except in disturbed soil close to the coal mine. Proximity to one of the industrial emitters was the main determinant of PAH and OPAH concentrations. The Sigma31PAHs concentrations correlated positively with the Sigma7 carbonyl-OPAH (r=0.98, p<0.01), Sigma5 hydroxyl-OPAH (r=…
Photo-polymerized lauryl methacrylate monolithic columns for CEC using lauroyl peroxide as initiator
2009
Lauryl methacrylate (LMA)-ester based monolithic columns photo-polymerized using lauroyl peroxide (LPO) as initiator were prepared, and their morphological and CEC properties were studied. The composition of the polymerization mixture (i.e. ratios of monomers/porogenic solvents, 1,4-butanediol/1-propanol and LMA/crosslinker) was optimized. The morphological and chromatographic properties of LMA columns were evaluated by means of SEM pictures and van Deemter plots of PAHs, respectively. The polymerization mixture selected as optimal provided a fast separation of a mixture of PAHs with excellent efficiencies (minimum plate heights of 8.9-11.1 μm). Satisfactory column-to-column (RS D < 4.5%) a…
CEC column behaviour of butyl and lauryl methacrylate monoliths prepared in non-aqueous media
2009
Polymeric monolithic stationary phases for capillary electrochromatography were prepared using two bulk monomers, butyl methacrylate (BMA) and lauryl methacrylate (LMA), by in situ polymerization in non-aqueous media. The effect of 1,4-butanediol/1-propanol ratio on porous properties was investigated separately for each monomer, keeping the proportion of monomers to pore-forming solvents fixed at 40:60 wt:wt. Also, mixtures of BMA and LMA at different 1,4-butanediol/1-propanol ratios were studied for tailoring the morphological features of the monolithic columns. The chromatographic performance of the different columns was evaluated by means of van Deemter plots of polycyclic aromatic hydro…
Retene, pyrene and phenanthrene cause distinct molecular-level changes in the cardiac tissue of rainbow trout (Oncorhynchus mykiss) larvae, part 1 – …
2020
Polycyclic aromatic hydrocarbons (PAHs) are contaminants of concern that impact every sphere of the environment. Despite several decades of research, their mechanisms of toxicity are still poorly understood. This study explores the mechanisms of cardiotoxicity of the three widespread model PAHs retene, pyrene and phenanthrene in the rainbow trout (Oncorhynchus mykiss) early life stages. Newly hatched larvae were exposed to each individual compound at sublethal doses causing no significant increase in the prevalence of deformities. Changes in the cardiac transcriptome were assessed after 1, 3, 7 and 14 days of exposure using custom Salmo salar microarrays. The highest number of differentiall…
Study of peak shape and efficiency in butyl acrylate-based monolithic columns for capillary electrochromatography.
2009
Abstract The study and modelling peak shape of in capillary electrochromatography (CEC), obtained using butyl acrylate (BA)-based monolithic columns, is described in this article. A modified-Gaussian model, which is a function of conventional experimental parameters: retention time (tR), peak height (H0) and standard deviation (σ0) at the peak maximum, and left (A) and right (B) halfwidths, was used to describe the peaks of thiourea and several polyaromatic hydrocarbons compounds, which were eluted under several applied voltages. A mean relative error below 2% in the prediction of peak shape is obtained. Based on peak parameters, an easy and reliable estimation of global chromatographic per…
Oxygen-containing polycyclic aromatic hydrocarbons (OPAHs) in urban soils of Bratislava, Slovakia: patterns, relation to PAHs and vertical distributi…
2011
Abstract We determined concentrations, sources, and vertical distribution of OPAHs and PAHs in soils of Bratislava. The ∑14 OPAHs concentrations in surface soil horizons ranged 88–2692 ng g−1 and those of ∑34 PAHs 842–244,870 ng g−1. The concentrations of the ∑9 carbonyl-OPAHs (r = 0.92, p = 0.0001) and the ∑5 hydroxyl-OPAHs (r = 0.73, p = 0.01) correlated significantly with ∑34 PAHs concentrations indicating the close association of OPAHs with parent-PAHs. OPAHs were quantitatively dominated by 9-fluorenone, 9,10-anthraquinone, 1-indanone and benzo[a]anthracene-7,12-dione. At several sites, individual carbonyl-OPAHs had higher concentrations than parent PAHs. The concentration ratios of se…
Chemical speciation of polycyclic aromatic hydrocarbons in sediments: partitioning and extraction of humic substances.
2010
The primary objective of this study was to examine the chemical speciation of polycyclic aromatic hydrocarbons (PAHs) found in sediments endowed with different characteristics. To achieve this purpose and because the nature of the sediments influences the distribution of PAHs, we have analyzed four different types of sediments. To study the role of organic matter in the sequestration of PAHs, we fractionated humic substances into humic acids and humin–mineral fractions. After their separation and purification, the humic components were examined for their sorptive reactivity by extracting them with organic solvents; these extracts were subsequently subjected to GC/MS analysis. Our results sh…
Polycyclic Aromatic Hydrocarbons in Sediments of Marine Coastal Lagoons in Messina, Italy: Extraction and GC/MS Analysis, Distribution and Sources
2004
The content of 21 polycyclic aromatic hydrocarbons (PAHs) has been determined in 16 samples of sediments collected from Ganzirri and Faro marine coastal lagoons in Messina (Italy). Analysis was performed by gas chromatography/mass spectrometry (GC/MS) in selected ion monitoring (SIM) mode. The total concentration of polycyclic aromatic hydrocarbons (PAHs) ranged from 74 to 5755 ...g/Kg of dry matrix. The resulting distributions and molecular ratios of specific compounds have been discussed in terms of sampling location and origin of organic matter. The results obtained show that levels of contamination are not homogeneous throughout the stations, while the relative distributions of PAH are…
Occurrence and removal of organic pollutants in sewages and landfill leachates
2002
Sewages of different composition and the effluents of four sewage treatment plants (STPs), plus sewage sludges were analysed for semivolatile organic priority pollutants. Furthermore, 11 landfill leachates were analysed to evaluate their contribution to sewage pollutants when co-treated. Bis(2-ethylhexyl) phthalate (DEHP) was the pollutant occurring at highest concentrations (up to 122 microg/l) and it was present in all sewages and leachates; concentrations of other phthalates were usually below 17 microg/l. Some polycyclic aromatic hydrocarbons (PAH) (<1 microg/l) and 2,6-dinitrotoluene (< or =5.9 microg/l) were also present in many of the sewages and leachates. Phthalates were present in…
