Arthrinins A–D: Novel diterpenoids and further constituents from the sponge derived fungus Arthrinium sp.

2011

Bioassay-guided fractionation of a methanolic extract of the fungus Arthrinium sp., isolated from the Mediterranean sponge Geodia cydonium, afforded 10 natural products including five new diterpenoids, arthrinins A-D (1-4) and myrocin D (5). In addition, five known compounds were obtained, which included myrocin A (6), norlichexanthone (7), anomalin A (8), decarboxycitrinone (9) and 2,5-dimethyl-7-hydroxychromone (10). The structures of all isolated compounds were unambiguously elucidated based on extensive 1D and 2D NMR and HR-MS analyzes. The absolute configuration of arthrinins A-D (1-4) was established by the convenient Mosher method performed in NMR tubes and by interpretation of the R…

Volatile organic compounds in truffle (Tuber magnatum Pico): Comparison of samples from different regions of Italy and from different seasons

2015

AbstractIn this paper volatile organic compounds (VOCs) from Tuber magnatum fruiting bodies were analyzed using a PTR-TOF-MS instrument. The aim was to characterize the VOC's profile of the fruiting bodies and identify if any VOCs were specific to a season and geographical areas. Multiple factorial analysis (MFA) was carried out on the signals obtained by MS. Experiments using ITS region sequencing proved that the T. magnatum life cycle includes the formation of fruiting bodies at two different times of the year. The VOCs profiles diverge when different seasonal and geographical productions are considered. Using PTR-TOF-MS, compounds present at levels as low pptv were detected. This made it…

Analysis of non-Saccharomyces yeast populations isolated from grape musts from Sicily (Italy).

2008

Aims The aim of this study was to identify the non-Saccharomyces yeast populations present in the grape must microflora from wineries from different areas around the island of Sicily. Methods and results Yeasts identification was conducted on 2575 colonies isolated from six musts, characterized using Wallerstein Laboratory (WL) nutrient agar, restriction analysis of the amplified 5.8S-internal transcribed spacer region and restriction profiles of amplified 26S rDNA. In those colonies, we identified 11 different yeast species originating from wine musts from two different geographical areas of the island of Sicily. Conclusions We isolated non-Saccharomyces yeasts and described the microflora…

Effects of hydrostatic pressure on yeasts isolated from deep-sea hydrothermal vents

2015

International audience; Hydrostatic pressure plays a significant role in the distribution of life in the biosphere. Knowledge of deep-sea piezotolerant and (hyper)piezophilic bacteria and archaea diversity has been well documented, along with their specific adaptations to cope with high hydrostatic pressure (HHP). Recent investigations of deep-sea microbial community compositions have shown unexpected micro-eukaryotic communities, mainly dominated by fungi. Molecular methods such as next-generation sequencing have been used for SSU rRNA gene sequencing to reveal fungal taxa. Currently, a difficult but fascinating challenge for marine mycologists is to create deep-sea marine fungus culture c…

Two new species of the genus Ochroconis, O. lascauxensis and O. anomala isolated from black stains in Lascaux Cave, France

2012

In the year 2001, some conspicuous black stains appeared on the walls of Lascaux Cave in France, which progressively disseminated throughout the cave. These black stains were so evident by 2007 that they have become one of the cave's major problems. In a mycological study of the black stains, Ochroconis strains were abundant among the isolates and constituted the major group of melanised fungi. Two new species of the genus Ochroconis, O. lascauxensis and O. anomala, were isolated and described. The description is based on the morphology of the fungi and the phylogenetic relationships of two of its gene regions internal transcribed spacer (ITS) and RNA polymerase II subunit B (RPB2). In addi…

Fungal biodiversity and in situ conservation in Italy

2011

A remarkable increase in knowledge of fungal biodiversity in Italy has occurred in the last five years. The authors report up-to-date numbers of fungi (Basidiomycota and Ascomycota) by regions together with distributional and ecological data on hypogeous fungi. Specific case studies such as alpine fungi, orchid mycorrhizas symbionts, invasive species, and the use of macrofungi as food by red squirrels are analyzed. In situ conservation strategies carried out on target species and/or taxonomic groups are also indicated.

The taxonomic position of Roesleria subterranea

2006

The genus Roesleria was introduced with the single species Roesleria hypogaea (current name: R. subterranea) by Thümen in 1877. The species was originally described from roots of grapevine and recognised as a facultative root-rotting parasite. The mazaediate ascoma with evanescent asci led to the assumption that Roesleria would be an ally of mazaediate lichens. In this study we calculate 28S, 18S as well ITS1-5.8S-ITS2 rDNA phylogenies. These data indicate that Roesleria is closely related to Hymenoscyphus (Helotiales). In contrast to other members of the Helotiales, the ascospores of Roesleria are passively released, and a distinct apical apparatus cannot be observed by LM. Electron optic …

Determination of the stability of protein pools from the cell wall of fungi.

2002

Stability of the protein populations present in the cell wall of three ascomycetous fungi Candida albicans, Saccharomyces cerevisiae and Yarrowia lipolytica was investigated. Cell wall proteins were either labeled with biotin or radiolabeled with amino acids, and chased for a period of time representing several generations. Proteins linked by non-covalent or covalent bonds were separated and their turnover was analyzed. No significant turnover took place during the chase period, and in fact radioactive proteins were accumulated in the wall during the period possibly by transfer through the secretory pathway. This transfer did not involve de novo protein synthesis; it was inhibited by azide,…

Molecular phylogeny and ultrastructure of the lichen microalga Asterochloris mediterranea sp. nov. from Mediterranean and Canary Islands ecosystems

2015

The microalgae of the genus Asterochloris are the preferential phycobionts in Cladonia, Lepraria and Stereocaulon lichens. Recent studies have highlighted the hidden diversity of the genus, even though phycobionts hosting Cladonia spp. in Mediterranean and Canarian ecosystems have been poorly explored. Phylogenetic analyses were made by concatenation of the sequences obtained with a plastid -LSU rDNA- and two nuclear -ITS rDNA and actin- molecular markers of the phycobionts living in several populations of Cladonia convoluta-C. foliacea complex, C. rangiformis and C. cervicornis species widely distributed in these areas in a great variety of substrata and habitats. A new strongly supported …

Air Quality Assessment by the Determination of Trace Elements in Lichens (Xanthoria calcicola) in an Industrial Area (Sicily, Italy)

2022

This study provides data on variation in the content of metals and metalloids measured in the lichens (Xanthoria calcicola Oxner) collected in the Syracusan petrochemical complex (Sicily, Italy) which is considered one of the largest in Europe. Concentrations of eighteen trace elements measured in the lichens that were collected from 49 different points were analyzed using an inductively coupled plasma (ICP-MS) device. The concentrations of the typical elements of industrial emissions (As, Cr, Ni, and V) highlight the environmental criticality that exists in the study area. The interpretation of the data in terms of multi-element statistical analysis (FA) and enrichment factor (EFs) proved …
