Cyanocobalamin Ultraflexible Lipid Vesicles: Characterization and In Vitro Evaluation of Drug-Skin Depth Profiles

2021

Atopic dermatitis (AD) and psoriasis are the most common chronic inflammatory skin disorders, which importantly affect the quality of life of patients who suffer them. Among other causes, nitric oxide has been reported as part of the triggering factors in the pathogenesis of both conditions. Cyanocobalamin (vitamin B12) has shown efficacy as a nitric oxide scavenger and some clinical trials have given positive outcomes in its use for treating skin pathologies. Passive skin diffusion is possible only for drugs with low molecular weights and intermediate lipophilicity. Unfortunately, the molecular weight and hydrophilicity of vitamin B12 do not predict its effective diffusion through the skin…

The Influence of Microbiome Dysbiosis and Bacterial Biofilms on Epidermal Barrier Function in Atopic Dermatitis—An Update

2021

Atopic dermatitis (AD) is a common inflammatory dermatosis affecting up to 30% of children and 10% of adults worldwide. AD is primarily driven by an epidermal barrier defect which triggers immune dysregulation within the skin. According to recent research such phenomena are closely related to the microbial dysbiosis of the skin. There is growing evidence that cutaneous microbiota and bacterial biofilms negatively affect skin barrier function, contributing to the onset and exacerbation of AD. This review summarizes the latest data on the mechanisms leading to microbiome dysbiosis and biofilm formation in AD, and the influence of these phenomena on skin barrier function.

Evaluation of Erythema Severity in Dermatoscopic Images of Canine Skin: Erythema Index Assessment and Image Sampling Reliability.

2021

The regular monitoring of erythema, one of the most important skin lesions in atopic (allergic) dogs, is essential for successful anti-allergic therapy. The smartphone-based dermatoscopy enables a convenient way to acquire quality images of erythematous skin. However, the image sampling to evaluate erythema severity is still done manually, introducing result variability. In this study, we investigated the correlation between the most popular erythema indices (EIs) and dermatologists’ erythema perception, and we measured intra- and inter-rater variability of the currently-used manual image-sampling methods (ISMs). We showed that the EIBRG, based on all three RGB (red, green, and blue) channe…

Selection of erythema index and image sampling method for the objective erythema estimation in dogs with atopic dermatitis

2020

Canine atopic dermatitis (CAD) is a common inflammatory and pruritic skin disease associated with allergy to exogenous allergens. The regular monitoring of skin lesions is essential to execute the anti-allergic therapy successfully. Erythema is one of the most important CAD-related lesions since it represents acute skin inflammation. Previously, we studied two optical systems (i.e., multispectral and dermatoscopic devices), which could objectively estimate erythema severity. However, we did not investigate, which image sampling method for selecting erythema-representing pixels and erythema index (EI) are correlated with the visual erythema assessment the most. In this paper, we tested three…

Vitamin D receptor gene polymorphisms in atopy

2021

Abstract Background The occurrence of allergic conditions, for example allergic asthma, rhinitis, and atopic dermatitis, is rising worldwide. These allergic conditions are associated with poor life quality. Vitamin D is proposed to be linked with increased risk and severe forms of allergic diseases. Aims This review article aimed to evaluate the vitamin D level role and polymorphisms of vitamin D receptor gene (VDR) in atopy. Methods & Materials We analyzed publications that were focusing on levels of vitamin D and/or polymorphism analysis of vitamin D receptor gene in allergic asthma, rhinitis, and atopic dermatitis patients. Results We noticed that levels of vitamin D are extensively stud…

Advances in antioxidant phytochemical for inflammatory skin diseases: mangiferin and naringin nanocarriers based lipids

2020

Chronic inflammatory skin diseases are the most common topical disorders and the fourth cause of global disability. Among all, psoriasis (PS) and atopic dermatitis (AD) are the most frequent chronic and recurrent inflammatory diseases of the skin. Atopic dermatitis has a strong Th2 factor while psoriasis is a disease driven by Th17. The actual treatment, which involve the combination of topical, systemic and biological therapy, is characterized by numerous adverse effects, which reduce patient compliance, lead to the limitation of long-term use and limit the therapeutic efficacy. Therefore, there is a great need for the continuous development of novel, effective and safe coadjuvant treatmen…

Characterization of disease-specific cellular abundance profiles of chronic inflammatory skin conditions from deconvolution of biopsy samples

2019

Background Psoriasis and atopic dermatitis are two inflammatory skin diseases with a high prevalence and a significant burden on the patients. Underlying molecular mechanisms include chronic inflammation and abnormal proliferation. However, the cell types contributing to these molecular mechanisms are much less understood. Recently, deconvolution methodologies have allowed the digital quantification of cell types in bulk tissue based on mRNA expression data from biopsies. Using these methods to study the cellular composition of the skin enables the rapid enumeration of multiple cell types, providing insight into the numerical changes of cell types associated with chronic inflammatory skin c…
