Nociceptin/orphanin FQ opioid receptor (NOP) selective ligand MCOPPB links anxiolytic and senolytic effects

2021

Accumulation of senescent cells may drive age-associated alterations and pathologies. Senolytics are promising therapeutics that can preferentially eliminate senescent cells. Here, we performed a high-throughput automatized screening (HTS) of the commercial LOPAC®Pfizer library on aphidicolin-induced senescent human fibroblasts, to identify novel senolytics. We discovered the nociceptin receptor FQ opioid receptor (NOP) selective ligand 1-[1-(1-methylcyclooctyl)-4-piperidinyl]-2-[(3R)-3-piperidinyl]-1H-benzimidazole (MCOPPB, a compound previously studied as potential anxiolytic) as the best scoring hit. The ability of MCOPPB to eliminate senescent cells in in vitro models was further tested…

Associations Between Lipoprotein Subfractions and Area and Density of Abdominal Muscle and Intermuscular Adipose Tissue: The Multi-Ethnic Study of At…

2021

Skeletal muscle quantity and quality decrease with older age, which is partly attributed to ectopic fat infiltration and has negative metabolic consequences. To inform efforts to preserve skeletal muscle with aging, a better understanding of biologic correlates of quantity and quality of muscle and intermuscular adipose tissue (IMAT) is needed. We used targeted lipidomics of lipoprotein subfractions among 947 Multi-Ethnic Study of Atherosclerosis participants to provide a detailed metabolic characterization of area and density of abdominal muscle and IMAT. Serum lipoprotein subfractions were measured at the first visit using 1H-Nuclear Magnetic Resonance spectroscopy. Muscle and IMAT area (…

LXR antagonists induce ABCD2 expression

2014

X-linked adrenoleukodystrophy (X-ALD) is a rare neurodegenerative disorder characterized by the accumulation of very-long-chain fatty acids resulting from a beta-oxidation defect. Oxidative stress and inflammation are also key components of the pathogenesis. X-ALD is caused by mutations in the ABCDI gene, which encodes for a peroxisomal half ABC transporter predicted to participate in the entry of VLCFA-CoA into the peroxisome, the unique site of their beta-oxidation. Two homologous peroxisomal ABC transporters, ABCD2 and ABCD3 have been proven to compensate for ABCD1 deficiency when overexpressed. Pharmacological induction of these target genes could therefore represent an alternative ther…

Proving convexity preserving properties of interpolatory subdivision schemes through reconstruction operators

2013

We introduce a new approach towards proving convexity preserving properties for interpolatory subdivision schemes. Our approach is based on the relation between subdivision schemes and prediction operators within Harten's framework for multiresolution, and hinges on certain convexity properties of the reconstruction operator associated to prediction. Our results allow us to recover certain known results [10,8,1,7]. In addition, we are able to determine the necessary conditions for convexity preservation of the family of subdivision schemes based on the Hermite interpolation considered in [4].

Nonlocalization Properties of Time Operators Transformations

2014

It is presented a general approach to the problem of extension of time operators and the associated Lambda transformations on singular measures. It is also shown that Lambda transformations defined on function spaces having the Urysohn property are non localized. Particular attention has been devoted to time and Lambda operators associated with the Walsh-Paley system and to a characterization of their domain and non locality.

An Integral Version of Ćirić’s Fixed Point Theorem

2011

We establish a new fixed point theorem for mappings satisfying a general contractive condition of integral type. The presented theorem generalizes the well known Ciric's fixed point theorem [Lj. B. Ciric, Generalized contractions and fixed point theorems, Publ. Inst. Math. 12 (26) (1971) 19-26]. Some examples and applications are given.

Comparative chemistry of 18-electron Mo(II) and 17-electron Mo(III) compounds containing only carbon-based ligands

1998

International audience; The chemical reactivity of various kinetically stable isomers of compound CpMo(η3-C3H5)(η4-C4H6), 1, and its oxidation product [1]+, as well as the bis-allyl Mo(III) complex CpMo(η3-C3H5)2, 2, and the bis-diene Mo(II) complex [CpMo(η4-C4H6)2]+, 3, is reviewed. The inertness toward isomerization processes of the allyl and butadiene ligands in the Mo(II) complexes has allowed a study of the relative reactivity toward both electrophilic and nucleophilic addition processes as a function of coordination mode. The dependence of various reaction pathways on the metal oxidation state has also been investigated. Of particular interest is the discovery that the electronically …

Regional variations in the use of complementary and alternative medicines for inflammatory bowel disease patients in Italy:an IG-IBD study.

2010

Abstract Background and aim: Complementary and alternative medicines (CAM) are being used increasingly by patients with Crohn's disease (CD) and ulcerative colitis (UC). We aimed to assess the prevalence and usage of CAM in different geographical areas of Italy and possible predictors of their use. Methods and materials: A structured questionnaire, administered to outpatients, attending 8 general hospitals and 9 tertiary referral centres, was completed by 2011 patients (909 CD, 1087 UC and 15 indeterminate colitis). 583 patients lived in the North, 659 in Central Italy and 769 in the South. Results: CAM users were 475 (23.6%) with no regional differences in their distribution. Usage correla…
