Subcellular distribution of ras in human and murine fibroblasts.

Abstract Ras proteins play a significant role in signal transduction in response to growth factors and in cell transformation. To be active, ras has to be translocated to the cell membrane. Since subcellular distribution has been mainly studied in vector-transformed cells which highly express ras proteins, and it has been difficult to detect ras in cells expressing the protein at physiological levels, we studied subcellular distribution in human and murine fibroblasts. Here we show for the first time that a significant amount of ras is associated with the membrane skeleton and the cytoskeleton.

El odontoameloblastoma (OA) es un tumor odontogénico mixto extremadamente raro que aparece en los huesos maxilares y que presenta tanto componentes epiteliales como mesenquimales. El término odontoameloblastoma fue incluido en la clasificación de 1971 de la OMS. Tan solo 23 casos bien documentados han sido publicados. Debido a su rareza, existe controversia en cuanto al tratamiento de este tumor. Cytotoxic effect of silorane and methacrylate based composites on the human dental pulp stem cells and fibroblasts

Objectives: The aim of this study was to compare the cytotoxic effect of a methacrylate-based and a silorane- based composite on the human dental pulp stem cells (DPSCs) versus human dental pulp fibroblasts (DPFs). Study Design: Samples of the Filtek Z250 and P90 were polymerized and immersed in the culture medium to obtain extracts after incubation for one, seven and 14 days. Magnetic cell sorting based on the CD146 expression was performed to purify DPSCs and DPFs. After incubation of both cells with the extracts, cytotoxicity was de - termined using the MTT test. Retinoblastoma is the most common intraocular malignancy afflicting children. The incidence is higher in developing countries, where treatment is limited and long-term survival rates are low. Microvesicles shed by oligodendroglioma cells and rheumatoid synovial fibroblasts contain aggrecanase activity

Membrane microvesicle shedding is an active process and occurs in viable cells with no signs of apoptosis or necrosis. We report here that microvesicles shed by oligodendroglioma cells contain an ‘aggrecanase’ activity, cleaving aggrecan at sites previously identified as targets for adamalysin metalloproteinases with disintegrin and thrombospondin domains (ADAMTSs). Degradation was inhibited by EDTA, the metalloproteinase inhibitor GM6001 and by tissue inhibitor of metalloproteinases (TIMP)-3, but not by TIMP-1 or TIMP-2. Sea urchin embryos are characterized by an extremely simple mode of development, rapid cleavage, high transparency, and well-defined cell lineage. Although they are not suitable for genetic studies, other approaches are successfully used to unravel mechanisms and molecules involved in cell fate specification and morphogenesis. Microinjection is the elective method to study gene function in sea urchin embryos.
