Amperometric detection of extended-spectrum β-lactamase activity : application to the characterization of resistant E.coli strains

2015

EA MERS CT3; International audience; The amperometric detection of extended-spectrum β-lactamase (ESBL) with carbon screen-printed sensors was investigated in the presence of the Nitrocefin, a commercially-available β-lactamase chromogenic cephalosporin substrate. Using an ESBL isolated from a clinical sample, it was shown for the first time that the intensity of a specific anodic pic current (EP = [similar]+0.3 V vs. Ag/AgCl) resulting from the catalytic hydrolysis of the β-lactam ring was proportional to the amount of ESBL. The proof-of-principle of a novel susceptibility assay for the rapid and accurate identification of ESBL- producing bacteria was then demonstrated. The detection schem…

The occurrence of extended-spectrum β-lactamase producing Shigella spp. in Tehran, Iran

2013

Background and Objectives: The emergence of extended-spectrum β-lactamase (ESBL)-producing Shigella spp. is of increasing clinical concern specially in children worldwide. The aim of this study was to investigate the occurrence of extended-spectrum β-lactamase producing Shigella spp. in Tehran, Iran. Materials and Methods: The study included all Shigella isolates recovered from pediatric patients aged less than 12 years admitted to a major pediatric hospital in Tehran, Iran, from 2008 to 2010. Bacterial identification, antimicrobial susceptibility testing, extended spectrum β-lactamases (ESBLs) screening and confirmatory tests were performed according to the standard guidelines. Conjugal tr…

Treated wastewaters discharged from wastewater treatment plants (WWTP) and livestock effluents are the main sources of contamination of aquatic environments and of agricultural soils by human and animal fecal micro-organisms. Contaminated waters, especially bathing waters, may present a sanitary risk for humans if the concentrations of fecal micro-organisms are high, thus indicating the potential presence of pathogenic and/or antibiotic resistant strains. Soils and crops also can be contaminated by these micro-organisms when treated wastewaters are used to irrigate cultivated soils. Consequently, microbiological quality controls are mandatory for bathing waters and treated wastewaters to ma…
