Two-Armed Activation of Bone Mineral Deposition by the Flavones Baicalin and Baicalein, Encapsulated in Polyphosphate Microparticles
2017
In this study, we investigated the effect of the two flavonoids, baicalin (baicalein 7-O-[Formula: see text]- d-glucuronic acid) and its aglycone, baicalein (5,6,7-trihydroxyflavone), after encapsulation into amorphous calcium polyphosphate (Ca-polyP) microparticles on mineralization of primary human osteoblasts (phOSB). Both flavonoids, which come from root extracts of Scutellaria baicalensis Georgi, are used in Traditional Chinese Medicine, and are nontoxic in cells up to a concentration of 3[Formula: see text][Formula: see text]g/ml. The morphogenetically active, energy-rich Ca-polyP particles with a stoichiometric P:Ca ratio of 1:2 are degraded by cellular alkaline phosphatase (ALP) to…
Negative pressure cavitation accelerated processing for extraction of main bioactive flavonoids from Radix Scutellariae
2011
Abstract To enhance the extraction efficiency and reduce the energy consumption, an emerging technology named negative pressure cavitation extraction (NPCE) has been shown to be a feasible option for the extraction of bioactive compounds in agricultural crops and medicinal plants. Meanwhile, it can be applied at the pilot scale as a manufacturing process for edible and medicinal plants. Currently, NPCE was proposed for extraction of baicalin, wogonoside, baicalein and wogonin from Radix Scutellariae on the basis of a central composite design (CCD) and response surface methodology (RSM). With proper optimization (80 mesh of particle size, 40 mL/g of liquid/solid ratio, 75% aqueous ethanol as…
Effects of Indole-3-Carbinol and Flavonoids Administered Separately and in Combination on Nitric Oxide Production and iNOS Expression in Rats
2010
Beneficial effects of natural compounds are often attributed to modulation of NO production; however effects produced by plant extracts do not correlate with effects of purified components. The goal of our work was to study ability of flavonoids and indole-3-carbinol, as well as their combinations to modify NO production, iNOS gene and protein expression in rat tissues. Baicalein and luteolin decreased NO concentration in both intact and LPS-treated animals. Baicalein decreased iNOS gene expression. Luteolin decreased NO production in the liver and heart and number of iNOS-positive cells in the liver of LPS-treated animals. Combination of the two substances did not decrease the NO synthesis…
Oxidation of baicalein by tyrosinase and by o-quinones
2023
Baicalein (5,6,7-trihydroxyflavone) has been previously described as an inhibitor of tyrosinase (Guo et al. Int. J. Biol. Macromol. 118 (2018) 57–68). However, long before this article was published this flavonoid had been shown to be a substrate of this enzyme and a catecholic cofactor partially abolishing the lag-phase during oxidation of l-tyrosine. Other compounds with a 1,2,3-triphenol moiety, such as pyrogallol, gallic acid and its esters are also oxidized by tyrosinase. Gallic acid was also shown to reduce tyrosinase-generated o-quinones. We have demonstrated that baicalein is also rapidly oxidized by o-quinones generated from catechols by tyrosinase or by treatment with sodium perio…
Rapid determination of baicalin and total baicalein content in Scutellariae radix by ATR-IR and NIR spectroscopy
2013
In this study methods for the quantification of baicalin and total baicalein in Scutellariae radix with near infrared (NIR) spectroscopy and attenuated-total-reflectance mid-infrared (ATR-IR) spectroscopy in hyphenation with multivariate analysis were developed and compared. The reference analysis was performed by high performance liquid chromatography coupled to diode array detection (HPLC-DAD). Different pretreatments like standard normal variate (SNV), multiplicative scatter correction (MSC), first and second derivative Savitzky-Golay were applied on the spectra to optimize the calibrations. A principal component analysis was performed with both spectroscopic methods to distinguish wild …
