Methods for studying root colonization by introduced beneficial bacteria

2003

e-Book; Some free-living rhizobacteria are considered as potential biocontrol and plant growth-promoting agents. Successful application of beneficial bacteria as microbial inoculants requires their presence and activity at the appropriate level, but even more, at the right time and place. Various markers are described in the literature to differentiate introduced bacteria from indigenous microflora and to visualize them. These markers are presented together with the methods currently applied to quantify bacterial densities and to characterize the distribution of introduced bacteria. The methods to quantify bacterial densities are either based on bacterial cultivation or not. Different types…

Costs-benfits trade-off in the intercation between Medicago truncatula and Pseudomonas fluorescens C7R12 across atmospheric carbon dioxide modulation

2011

The interactions between plants and soil microorganisms are mainly based on trophic relationships. The compounds exchanged represent cost for the organism produces them but a benefit for the one who receive those compounds. A mutualistic relation leads to stability in the cost-benefit balance resulting from a co-evolution between the two organisms. The cost corresponding for the release of carbon compounds by the plant would be offset by benefits in return corresponding for the activities of microorganisms that use them. We tested by an experimental way the effect of CO2 concentration on the interaction between M. truncatula and the bacterium P. fluorescens C7R12. The results allowed a best…

Soil as support of biodiversity and functions

2015

Chapitre 11 Section 3 EA SPE EcolDur GenoSol; The soil is a major reservoir of biological diversity on our planet. It also shelters numerous biological and ecological processes and therefore contributes to the production of a considerable number of ecosystem services. Among the ecological, social and economic services identified, the role of soil as a reservoir of diversity has now been well established, along with its role in nutrient cycling, supporting primary productivity, pollution removal and storing carbon. Since the development of industrialization, urbanization and agriculture, soils have been subjected to numerous variations in environmental conditions, which have resulted in modi…

Resveratrol from vine to wine

2013

SPE IPM Biochemistry Research Trends Chapitre I; The stilbene compound resveratrol and its derivatives are produced by various plant families and possess a wide spectrum of interesting medical properties. In Vitaceae, these compounds are phytoalexins ie antifungal metabolites synthesized and accumulated in response to biotic or abiotic stresses. Therefore, even if their level in wines is influenced by winemaking process and fermentative micro-organisms, it largely depends on their initial level in berries. Resveratrol is synthesized via the phenylalanine / polymalonate pathway and can undergo glycosylation, methylation or polymerization, leading to a set of derivatives. Due to their distinc…

Impact of stresses associated to climate changes on natural and induced grapevine resistance to pathogens

2013

France is a worldwide leader in grape production and has to face global changes including climate evolution and transitions towards sustainable systems of production. One major problem in viticulture is indeed the high susceptibility of most of the grown cultivars to cryptogamic diseases, such as downy mildew (Plasmopara viticola). Maintaining a satisfactory health status of grapevine currently requires numerous fungicide treatments. However, in an objective of sustainable viticulture, there is increasing societal request, winegrower awareness, and political incitation to reduce this use of pesticides. For these reasons, alternative strategies of protection are currently investigated at the…

Stilbenes and resveratrol metabolites improve mitochondrial fatty acid oxidation defects in human fibroblasts

2014

International audience; Background: Inborn enzyme defects of mitochondrial fatty acid beta-oxidation (FAO) form a large group of genetic disorders associated to variable clinical presentations ranging from life-threatening pediatric manifestations up to milder late onset phenotypes, including myopathy. Very few candidate drugs have been identified in this group of disorders. Resveratrol (RSV) is a natural polyphenol with anti-oxidant and anti-inflammatory effects, recently shown to have beneficial metabolic properties in mice models. Our study explores its possible effects on FAO and mitochondrial energy metabolism in human cells, which are still very little documented. Methods: Using cells …

Dual mode of action of grape cane extracts against Botrytis cinerea

2019

International audience; Crude extracts of Vitis vinifera canes represent a natural source of stilbene compounds with well characterized antifungals properties. In our trials, exogenous application of a stilbene extract (SE) obtained from grape canes on grapevine leaves reduces the necrotic lesions caused by Botrytis cinerea. The SE showed to possess a direct antifungal activity by inhibiting the mycelium growth. The activation of some grapevine defense mechanism was also investigated. H2O2 production and activation of mitogen-activated protein kinase (MAPK) phosphorylation cascades as well as accumulation of stilbenoid phytoalexins were explored on grapevine cell suspension. Moreover, the t…
