4-4-20 anti-fluorescyl IgG Fab' recognition of membrane bound hapten: direct evidence for the role of protein and interfacial structure.

1995

The surface forces apparatus was used to identify the molecular forces that control the interactions of monoclonal 4-4-20 antifluorescyl IgG Fab' fragments with fluorescein-presenting supported planar bilayers. At long range, the electrostatic force between oriented Fab' and fluorescein monolayers was controlled by the composition of the protein exterior surrounding the antigen-combining site rather than by the overall protein charge. The measured positive electrostatic potential of the Fab' monolayer at pH > pI(Fab') was consistent with the structure of the exposed Fab' surface in which a ring of positive charge at the mouth of the antigen-combining site dominates the local electrostatic s…

Static dielectric constant, viscosity, and structure of pure isomeric pentanols

1981

Static dielectric constants, viscosities, densitites and refractive indices of 2-pentanol, 3-pentanol, 2-methyl-1-butanol, 3-methyl-1-butanol and 2-methyl-2-butanol were measured at 15, 25, 35 and 45°C. These results together with the previous data on n-pentanol have been analyzed in terms of the Kirkwood correlation factor g k and of the energy of activiation for viscous flow. With the exception of 2-methyl-2-butanol, g k was found to be greater than unity. These results show that the monomeric units of isomeric pentanols interact by means of hydrogen bonding to form dynamic structures essentially of two types: linear chains where co-association raises the total polarizability and cyclic d…

Molecular association in water-isomeric pentanol mixtures at 25C

1982

The results of static dielectric constant and viscosity measurements on solutions of water (concentration range 0 to 0.3 mole fraction) in 2-pentanol, 3-pentanol, 2-methyl-1-butanol, 3-methyl-1-butanol, and 2-methyl-2-butanol together with previous results for solutions of water in n-pentanol are discussed in terms of the information that they provide on the nature and the extent of molecular association in these solutions. We conclude that in most systems this association leads to the formation of tetrahedral complexes such as H2O(ROH)4. Evidences of the correlation of water-alcohol interactions with molecular parameters (position of OH group, steric hindrance of alkyl chain) of alcohols a…

Recent Probes of Standard and Non-standard Neutrino Physics With Nuclei

2019

We review standard and non-standard neutrino physics probes that are based on nuclear measurements. We pay special attention on the discussion of prospects to extract new physics at prominent rare event measurements looking for neutrino-nucleus scattering, such as the coherent elastic neutrino-nucleus scattering (CE$\nu$NS) that may involve lepton flavor violation (LFV) in neutral-currents (NC). For the latter processes several appreciably sensitive experiments are currently pursued or have been planed to operate in the near future, like the COHERENT, CONUS, CONNIE, MINER, TEXONO, RED100, vGEN, Ricochet, NUCLEUS etc. We provide a thorough discussion on phenomenological and theoretical studi…

Hadamard NMR imaging with slice selection

1996

Stochastic NMR imaging is one of the less common NMR imaging techniques. Nevertheless, stochastic rf excitation is characterized by some remarkable features: the rf excitation power is at least two orders of magnitude lower in comparison to conventionally pulsed NMR imaging schemes. Thus, the technique is of interest for imaging of large objects. The systematic noise inherent in images obtained with random noise excitation has been eliminated by using pseudorandom noise together with Hadamard transformation for data evaluation. Data acquisition times are comparable to those of ultrafast imaging techniques. For slice selection, z magnetization is destroyed outside the slice region with speci…

Evidence of stochastic resonance in the mating behavior of Nezara viridula (L.)

2008

We investigate the role of the noise in the mating behavior between individuals of Nezara viridula (L.), by analyzing the temporal and spectral features of the non-pulsed type female calling song emitted by single individuals. We have measured the threshold level for the signal detection, by performing experiments with the calling signal at different intensities and analyzing the insect response by directionality tests performed on a group of male individuals. By using a sub-threshold signal and an acoustic Gaussian noise source, we have investigated the insect response for different levels of noise, finding behavioral activation for suitable noise intensities. In particular, the percentage…

Recent advances in surface plasmon resonance biosensors for microRNAs detection

2020

miRNAs are a large family of non-coding RNAs which play important roles in translational and post-transcriptional regulation of gene expression and biological processes. Abnormal expression of miRNAs is related to the initiation and progression of different diseases which make them be promising candidates for early medical diagnostics. Thus, accurate detection of miRNAs has great significance for disorder diagnosis. Nevertheless, their intrinsic characteristics such as short sequence, low concentration and sequence homology challenge routine techniques. The detection assays need to be extremely sensitive and selective in small value of intricate RNA samples. Biosensor-based strategies have …

High-resolution crystal structure of an avidin-related protein: insight into high-affinity biotin binding and protein stability.

2004

The chicken avidin gene belongs to an extended gene family encoding seven avidin-related genes (AVRs), of which only avidin is expressed in the chicken. The sequences of AVR4 and AVR5 are identical and the common protein (AVR4) has been expressed both in insect and bacterial systems. The recombinant proteins are similarly hyperthermostable and bind biotin with similarly high affinities. AVR4 was crystallized in the apo and biotin-complexed forms and their structures were determined at high resolution. Its tertiary and quaternary structures are very similar to those of avidin and streptavidin. Its biotin-binding site shows only a few alterations compared with those of avidin and streptavidin…

Specific interaction of desthiobiotin lipids and water-soluble biotin compounds with streptavidin

1991

As shown for biotin lipids (Ref. 1), the formation of perfect 2-D crystalline streptavidin domains can also be observed in the plane of desthiobiotin lipid monolayers. The binding constant of streptavidin with desthiobiotin (Ka = 5·1013 mol−1) is lower than that with biotin (Ka = 1015 mol−1) (Ref. 2). By adding free biotin into the subphase a competitive replacement and a detaching of the streptavidin domains from the desthiobiotin lipid monolayer takes place. Streptavidin domains built at receptor lipid monolayers are still functional. As could be shown, there are two biotin binding sites at each protein molecule that are fully accessible to biotin (Ref. 1). This can be proven by the inter…

Production of Hev b5 as a fluorescent biotin-binding tripartite fusion protein in insect cells

2005

The presented green fluorescent protein and streptavidin core-based tripartite fusion system provides a simple and efficient way for the production of proteins fused to it in insect cells. This fusion protein forms a unique tag, which serves as a multipurpose device enabling easy optimization of production, one-step purification via streptavidin-biotin interaction, and visualization of the fusion protein during downstream processing and in applications. In the present study, we demonstrate the successful production, purification, and detection of a natural rubber latex allergen Hev b5 with this system. We also describe the production of another NRL allergen with the system, Hev b1, which fo…
