Paxillin is a potential participant in the direct intracellular force transmission which is considered as the foundation of cells sensing and responding to extracellular environment. However, the detection of tension across paxillin has not been achieved due to lacking microsized tools. Herein, a paxillin tension sensor (PaxTs) based on Fluorescence Resonance Energy Transfer (FRET) technique was constructed. PaxTs can be expressed and assembled to FA sites spontaneously to visualize the tension across paxillin with FRET efficiency of ~62.4% in living cells. Tämä Pro gradu -tutkielma koostuu kirjallisuuskatsauksesta ja suoritetun erikoistyön selostuksesta. Puhdistamattoman vuorisuolan, jota kutsutaan myös kristallisuolaksi tai Ruususuolaksi, käyttö on lisääntynyt runsaasti viime vuosina. Käyttöä perustellaan sen sisältämillä monilla alkuaineilla, mutta analyyttisiä tutkimuksia eri aineiden pitoisuuksista löytyy kirjallisuudesta niukasti. The purpose of the study was to examine the validity of core action elements and feeling states in ice hockey players in the prediction of performance. A second aim of the study was to explore the effectiveness of a 30-day program targeting action and emotion regulation. Participants were male ice hockey players drawn from two teams competing at the highest level of the junior Finnish ice hockey league. They were assigned to a self-regulation (n = 24) and a control (n = 19) group. The purpose of the current study aimed to examine the performance related experiences of elite karate athletes prior to a major international competition. The secondary aim was to explore the relationship between athlete and coach performance ratings. Athletes selected emotional and non-emotional words to describe their experiences related to performance. Results indicated that best performance was characterized by high intensities of helpful emotion and non-emotion descriptors, while worst performance was characterized by high intensities of harmful descriptors. Using the multi-states (MuSt) theory as a framework, the present study focused on the individual (personality) antecedents of psychobiosocial states. Psychobiosocial states comprise emotional subjective experiences and their correlates (cognitive, motivational, volitional, bodily, motor-behavioural, operational, and communicative), that can be functional for performance (helpful) or dysfunctional (harmful).
