GLUCAGON-LIKE PEPTIDE-1 MODULATES EXCITATORY CHOLINERGIC NEUROTRANSMISSION OF MOUSE DUODENUM AND COLON

2009

The incretin Glucagon-like-peptide-1 (GLP-1) is produced by enterendocrine L cells of the small intestine. It delays gastric emptying and reduces intestinal transit by inhibiting vagal activity. To date, no evidences are available about a direct peripheral influence on gastrointestinal smooth muscle cells. The purpose of the present study was to investigate in vitro the effects of GLP-1 on the spontaneous and evoked mechanical activity of murine duodenum and colon, recording intraluminal pressure, index of the circular mechanical activity, and isometric tension, index of the longitudinal mechanical activity. In both intestinal segments GLP-1 (up to 1 μM) did not affect spontaneous mechanica…

Amplification, contraction and genomic spread of a satellite DNA family (E180) in Medicago (Fabaceae) and allied genera

2011

†Background and Aims Satellite DNA is a genomic component present in virtually all eukaryotic organisms. The turnover of highly repetitive satellite DNA is an important element in genome organization and evolution in plants. Here we assess the presence and physical distribution of the repetitive DNA E180 family in Medicago and allied genera. Our goals were to gain insight into the karyotype evolution of Medicago using satellite DNA markers, and to evaluate the taxonomic and phylogenetic signal of a satellite DNA family in a genus hypothesized to have a complex evolutionary history. †Methods Seventy accessions from Medicago, Trigonella, Melilotus and Trifolium were analysed by PCR to assess …

Encapsulation of xenon by bridged resorcinarene cages with high 129Xe NMR chemical shift and efficient exchange dynamics

2023

Functionalized cages encapsulating xenon atoms enable highly sensitive, background-free molecular imaging through a technique known as HyperCEST 129Xe MRI. Here, we introduce a class of potential biosensor cage structures based on two resorcinarene macrocycles bridged either by aliphatic carbon chains or piperazines. First-principles-based modeling predicts a high chemical shift (about 345 ppm) outside the typical experimental observation window for 129Xe encapsulated by the aliphatically bridged cage and two 129Xe resonances for the piperazine-bridged cages corresponding to single and double loading. Based on the computational predictions as well as 129Xe chemical exchange saturation trans…

Detection of Hepatitis B Virus DNA in the Liver of Children with Chronic Hepatitis B by In Situ Hybridization and Its Relation to Other Viral Markers

1992

The aim of the study was to detect hepatitis B virus (HBV) DNA by in situ hybridization (ISH) with a 35S-labeled radioactive probe in frozen liver biopsy tissue sections of 63 hepatitis B virus surface antigen (HBsAg)-positive children. The results were compared to other markers of viral replication. HBV DNA was detected in 48 children. Of the 15 negative cases, four had hepatitis B envelope antigen (HBeAg), 10 anti-HBe, and one neither HBeAg nor anti-HBe. Free HBV DNA in serum and liver was positive in one patient. Forty of the positive children were HBeAg- and six anti-HBe-positive; two were negative for both. Of 45 36 had HBV DNA in serum. In 38 of 47 HBV DNA and in 31 of 42 HBcAg could …

Partial Sequence Homogenization in the 5S Multigene Families May Generate Sequence Chimeras and Spurious Results in Phylogenetic Reconstructions

2014

Multigene families have provided opportunities for evolutionary biologists to assess molecular evolution processes and phylogenetic reconstructions at deep and shallow systematic levels. However, the use of these markers is not free of technical and analytical challenges. Many evolutionary studies that used the nuclear 5S rDNA gene family rarely used contiguous 5S coding sequences due to the routine use of head-to-tail polymerase chain reaction primers that are anchored to the coding region. Moreover, the 5S coding sequences have been concatenated with independent, adjacent gene units in many studies, creating simulated chimeric genes as the raw data for evolutionary analysis. This practice…

Is the Secret in the Gut? SuperJump Activity Improves Bone Remodeling and Glucose Homeostasis by GLP-1 and GIP Peptides in Eumenorrheic Women

2022

We showed that twenty weeks of SuperJump activity, an innovative workout training performed on an elastic minitrampoline, reduced bone resorption and increased bone formation in eumenorrheic women acting on the key points of the regulation of bone metabolism. The present study analyzed whether the gastrointestinal hormones are involved in the mechanism of action and if it has an impact on glucose homeostasis. The control group was composed of twelve women, similar to the exercise group that performed SuperJump activity for twenty weeks. The analysis was performed on blood samples and investigated GLP-1, GIP, GLP-2, PYY, ghrelin, glucose, insulin, insulin resistance, β-cell function, and ins…
