[1] We report 40Ar/39Ar ages from various tectonic units in the Aegean and westernmost Turkey. On the basis of published geochronologic data and our 40Ar/39Ar ages we propose that the Aegean is made up of several high-pressure units, which were successively underplated from the Late Cretaceous until the Miocene. Ages for high-pressure metamorphism range from 80–83 Ma in parts of the Vardar-Izmir-Ankara suture zone in the north to 21–24 Ma for the Basal unit in the Cyclades and the external high-pressure belt on Crete in the south. Staphylococcus aureus clonal complex 398 (CC398) isolates cluster into two distinct phylogenetic clades based on single-nucleotide polymorphisms (SNPs) revealing a basal human clade and a more derived livestock clade. The scn and tet(M) genes are strongly associated with the human and the livestock clade, respectively, due to loss and acquisition of mobile genetic elements. We present canonical single-nucleotide polymorphism (canSNP) assays that differentiate the two major host-associated S. aureus CC398 clades and a duplex PCR assay for detection of scn and tet (M). An extensive screening of the human neuronal nitric oxide synthase (nNOS) mRNAs in various human tissues and cell lines unraveled an extreme complexity in the transcription of this gene. Using 5'rapid amplification of cDNA ends (5'-RACE), ten different exons 1 (named 1a-1l) were identified. They were spliced in a cell-specific manner to a common exon 2, which bears the translational start site. Three first exons (1 d, 1g and 1f) were used predominantly for the transcription of the nNOS gene (146 out of 197 5'-RACE clones contained these exons). In the rat pineal gland cAMP mediates nocturnal induction of the enzyme arylalkylamine N-acetyltransferase (AA-NAT) as well as of transcription factors such as inducible cAMP early repressor (ICER), Fos-related antigen-2 (Fra-2) and JunB. Cyclic AMP stimulates the phosphorylation of the DNA binding protein cAMP response element binding protein (CREB). While cAMP-induced CREB phosphorylation appears to be a prerequisite for AA-NAT and ICER gene expression, it is not known whether CREB phosphorylation accounts for the full cAMP response of the two genes. Expressional dysregulation of the human neuronal nitric oxide synthase (NOS1) gene represents an important mechanism in the pathogenesis of certain neuronal disease states. The structure and regulation of the human NOS1 gene is highly complex based on cell type- and stimulus-dependent usage of multiple exon 1 variants. Here we demonstrate that the untranslated region of exon 2 exerts promoter and enhancer functions as well, facilitated in large part by cooperative interaction of two conserved adjacent CREB/AP-1 binding sites. AbstractThe Serbo-Macedonian Massif is a basement complex in the Internal Hellenides of northern Greece, situated between the Vardar Zone to the west and the Rhodope Massif to the east. The Serbo-Macedonian Massif comprises several distinct basement units interpreted as terranes, the largest of which is the Gondwana-derived Vertiskos Terrane in the northwestern and central parts of the massif. A series of leucocratic meta-granites intrude the Silurian orthogneiss basement of the Vertiskos Terrane. No similar granites are found in any of the other units of the Internal Hellenides. Metagabbros in the core series of the Menderes Massif, for some time considered as post orogenic Miocene intrusives, revealed a strong poly-metamorphic history. The metagabbro bodies exhibit a pronounced zonation. Within their cores, igneous minerals are still preserved. Coronitic textures are interpreted as results of a high temperature, possibly granulitic overprint. The outer parts of the metagabbro bodies mostly consist of strongly to completely retrograded gamet-amphibo-lites, but occasionally contain relies of eclogites. The effects of adrenaline (2.2×10−6 M) and cyclic N6-2′-O-dibutyryl-adenosine-3′,5′-monophosphate (DB-c-AMP; 10−3 M) on mechanical performance, 45Ca uptake and total tissue calcium concentration were investigated in electrically stimulated left auricles isolated from female rats weighing 180–220 g. The experiments were performed at reduced [Ca]e of 0.45 mM and at various frequencies of stimulation (0–120 beats/min). In the first series of experiments 45Ca incubation time was 5 min. The effect of dibutyryl cyclic AMP (DB-c-AMP; 3 X 10(-4)-3 X 10(-3) M) on electrically induced twitch and high potassium (142.4 mM KCl)-induced contracture tension was studied in papillary muscles from normal and reserpinized cats ([Ca]0 1.8 mM; 25 degrees C; pH 7.4). In both groups of preparations, the increase in twitch tension evoked by DB-c-AMP was accompanied by an abbreviation of the time to peak force and of relaxation time. In the same preparations, the high potassium contracture was markedly depressed by DB-c-AMP in a concentration-dependent manner.
