Somatic copy number alterations are associated with EGFR amplification and shortened survival in patients with primary glioblastoma.
2019
Glioblastoma (GBM) is the most common malignant primary tumor of the central nervous system. With no effective therapy, the prognosis for patients is terrible poor. It is highly heterogeneous and EGFR amplification is its most frequent molecular alteration. In this light, we aimed to examine the genetic heterogeneity of GBM and to correlate it with the clinical characteristics of the patients. For that purpose, we analyzed the status of EGFR and the somatic copy number alterations (CNAs) of a set of tumor suppressor genes and oncogenes. Thus, we found GBMs with high level of EGFR amplification, low level and with no EGFR amplification. Highly amplified tumors showed histological features of…
Distinct Activities of Glycolytic Enzymes Identify Chronic Lymphocytic Leukemia Patients with a more Aggressive Course and Resistance to Chemo-Immuno…
2018
A higher capacity to grow under hypoxic conditions can lead to a more aggressive behavior of tumor cells. Determining tumor activity under hypoxia may identify chronic lymphocytic leukemia (CLL) with aggressive clinical course and predict response to chemo-immunotherapy (CIT). A metabolic score was generated by determining pyruvate kinase and lactate dehydrogenase, key enzymes of glycolysis, ex vivo in primary CLL samples under normoxic and hypoxic conditions. This score was further correlated with clinical endpoints and response to CIT in 96 CLL patients. 45 patients were classified as metabolic high risk (HR), 51 as low risk (LR). Treatment-free survival (TFS) was significantly shorter in…
The monocytic population in chronic lymphocytic leukemia shows altered composition and deregulation of genes involved in phagocytosis and inflammatio…
2013
Macrophages reside in tissues infiltrated by chronic lymphocytic leukemia B cells and the extent of infiltration is associated with adverse prognostic factors. We studied blood monocyte population by flow cytometry and whole-genome microarrays. A mixed lymphocyte reaction was performed to evaluate proliferation of T cells in contact with monocytes from patients and normal donors. Migration and gene modulation in normal monocytes cultured with CLL cells were also evaluated. The absolute number of monocytes increased in chronic lymphocytic leukemia patients compared to the number in normal controls (792 +/- 86 cells/mu L versus 485 +/- 46 cells/mL, P=0.003). Higher numbers of non-classical CD…
Analysis of chronic lymphotic leukemia transcriptomic profile: differences between molecular subgroups
2009
B cell chronic lymphocytic leukemia (CLL) is a lymphoproliferative disorder with a variable clinical course. Patients with unmutated IgV(H) gene show a shorter progression-free and overall survival than patients with immunoglobulin heavy chain variable regions (IgV(H)) gene mutated. In addition, BCL6 mutations identify a subgroup of patients with high risk of progression. Gene expression was analysed in 36 early-stage patients using high-density microarrays. Around 150 genes differentially expressed were found according to IgV(H) mutations, whereas no difference was found according to BCL6 mutations. Functional profiling methods allowed us to distinguish KEGG and gene ontology terms showing…
A novel role of the CX3CR1/CX3CL1 system in the cross-talk between chronic lymphocytic leukemia cells and tumor microenvironment
2011
Several chemokines/chemokine receptors such as CCR7, CXCR4 and CXCR5 attract chronic lymphocytic leukemia (CLL) cells to specific microenvironments. Here we have investigated whether the CX(3)CR1/CX(3)CL1 axis is involved in the interaction of CLL with their microenvironment. CLL cells from 52 patients expressed surface CX(3)CR1 and CX(3)CL1 and released constitutively soluble CX(3)CL1. One third of these were attracted in vitro by soluble CX(3)CL1. CX(3)CL1-induced phosphorylation of PI3K, Erk1/2, p38, Akt and Src was involved in induction of CLL chemotaxis. Leukemic B cells upregulated CXCR4 upon incubation with CX(3)CL1 and this was paralleled by increased chemotaxis to CXCL12. Akt phosp…
Development and application of an alternative modelling approach for the thermo-mechanical analysis of a DEMO water-cooled lithium lead breeding blan…
2022
In the frame of the EUROfusion research activities devoted to the design of the DEMO breeding blanket (BB), the Water-Cooled Lithium-Lead BB (WCLL) concept is one of the candidates currently assessed in EU. To this end, an intense research campaign is ongoing to develop a robust geometric configuration for the WCLL BB Central Outboard Segment (COB). Since the current reference design of the WCLL COB segment is not mature enough to allow a full thermal-hydraulic assessment, an alternative procedure aimed at obtaining a thermal field for the whole segment without performing its complete thermal-hydraulic analysis is presented and applied in this work. The scope of the work is to obtain a ther…
Tokamak cooling systems and power conversion system options
2022
DEMO will be a fusion power plant demonstrating the integration into the grid architecture of an electric utility grid. The design of the power conversion chain is of particular importance, as it must adequately account for the specifics of nuclear fusion on the generation side and ensure compatibility with the electric utility grid at all times. One of the special challenges the foreseen pulsed operation, which affects the operation of the entire heat transport chain. This requires a time-dependant analysis of different concept design approaches to ensure proof of reliable operation and efficiency to obtain nuclear licensing. Several architectures of Balance of Plant were conceived and dev…
Recent progress in developing a feasible and integrated conceptual design of the WCLL BB in EUROfusion project
2019
The water-cooled lithium-lead breeding blanket is in the pre-conceptual design phase. It is a candidate option for European DEMO nuclear fusion reactor. This breeding blanket concept relies on the liquid lithium-lead as breeder-multiplier, pressurized water as coolant and EUROFER as structural material. Current design is based on DEMO 2017 specifications. Two separate water systems are in charge of cooling the first wall and the breeding zone: thermo-dynamic cycle is 295–328 °C at 15.5 MPa. The breeder enters and exits from the breeding zone at 330 °C. Cornerstones of the design are the single module segment approach and the water manifold between the breeding blanket box and the back suppo…
CD38/CD31, the CCL3 and CCL4 chemokines, and CD49d/vascular cell adhesion molecule-1 are interchained by sequential events sustaining chronic lymphoc…
2009
AbstractCD38 and CD49d are associated negative prognosticators in chronic lymphocytic leukemia (CLL). Despite evidence that both molecules are involved in interactions occurring between CLL and normal cells in the context of CLL-involved tissues, a functional link is still missing. Using gene expression profiles comparing CD38+CD49d+ versus CD38−CD49d− CLL cells, we showed overexpression of the CCL3 and CCL4 chemokines in cells from the former group. These chemokines were also up-regulated by CD38 signals in CLL; moreover, CCL3 was expressed by CLL cells from bone marrow biopsies (BMB) of CD38+CD49d+ but not CD38−CD49d− cases. High levels of CCR1 and, to a lesser extent, CCR5, the receptors…
