Differentiation capacity of epithelial cells in the sponge Suberites domuncula.

2004

Sponges (phylum Porifera) represent the oldest metazoans. Their characteristic metazoan adhesion molecules and transcription factors enable them to establish a complex "Bauplan" ; three major differentiated cell types (epithelial cells, skeletal cells/sclerocytes, and contractile cells) can be distinguished. Since no molecular markers are as yet available to distinguish these somatic cells or the corresponding embryonic cells from which they originate, we have selected the following three genes for their characterization: noggin (a signaling molecule in development), a caspase that encodes an apoptotic molecule, and silicatein. Silicatein is an enzyme that is involved in the synthesis of si…

Histone H3 Lysine 4 Mono-methylation does not Require Ubiquitination of Histone H2B

2005

The yeast Set1-complex catalyzes histone H3 lysine 4 (H3K4) methylation. Using N-terminal Edman sequencing, we determined that 50% of H3K4 is methylated and consists of roughly equal amounts of mono, di and tri-methylated H3K4. We further show that loss of either Paf1 of the Paf1 elongation complex, or ubiquitination of histone H2B, has only a modest effect on bulk histone mono-methylation at H3K4. Despite the fact that Set1 recruitment decreases in paf1delta cells, loss of Paf1 results in an increase of H3K4 mono-methylation at the 5' coding region of active genes, suggesting a Paf1-independent targeting of Set1. In contrast to Paf1 inactivation, deleting RTF1 affects H3K4 mono-methylation…

Glutathione and cellular redox control in epigenetic regulation.

2015

Epigenetics is defined as the mitotically/meiotically heritable changes in gene expression that are not due to changes in the primary DNA sequence. Over recent years, growing evidence has suggested a link between redox metabolism and the control of epigenetic mechanisms. The effect of the redox control, oxidative stress, and glutathione (GSH) on the epigenetic mechanisms occur at different levels affecting DNA methylation, miRNAs expression, and histone post-translational modifications (PTMs). Furthermore, a number of redox PTMs are being described, so enriching the histone code. Pioneer works showed how oxidized GSH inhibits the activity of S-adenosyl methionine synthetase, MAT1A, a key en…

A chromatin-associated protein from pea seeds preferentially binds histones H3 and H4

2002

Pisum sativum p16 is a protein present in the chromatin of ungerminated embryonic axes. The purification of p16 and the isolation of a cDNA clone of psp54, the gene encoding its precursor have been recently reported [Castillo, J., Rodrigo, M. I., Marquez, J. A., Zuniga, A and Franco, L. (2000) Eur. J. Biochem.267, 2156-2165]. In the present paper, we present data showing that p16 is a nuclear protein. First, after subcellular fractionation, p16 was clearly found in nuclei, although the protein is also present in other organelles. Immunocytochemical methods also confirm the above results. p16 seems to be firmly anchored to chromatin, as only extensive DNase I digestion of nuclei allows its r…

Finite-range separable pairing interaction within the new N3LO DFT approach

2011

For over four decades, the Skyrme functional within various parametrizations has been used to calculate nuclear properties. In the last few years there was a number of attempts to improve its performance and introduce generalized forms. In particular, the most general phenomenological quasi-local energy density functional, which contains all combinations of density, spin-density, and their derivatives up to the sixth order (N3LO), was proposed in [1]. Since in the phenomenological-functional approaches, the particle-particle (pp) channel is treated independently of the particle-hole (ph) channel, there remains a question of what pairing interaction is suitable to use within the N3LO energy …

Analysis of a Station Black-Out transient in SMR by using the TRACE and RELAP5 code

2014

The present paper deals with the investigation of the evolution and consequences of a Station Black-Out (SBO) initiating event transient in the SPES3 facility [1]. This facility is an integral simulator of a small modular reactor being built at the SIET laboratories, in the framework of the R&D program on nuclear fission funded by the Italian Ministry of Economic Development and led by ENEA. The SBO transient will be simulated by using the RELAP5 and TRACE nodalizations of the SPES3 facility. Moreover, the analysis will contribute to study the differences on the code predictions considering the different modelling approach with one and/or three-dimensional components and to compare the capa…

RELAP5-3D thermal hydraulic analysis of the target cooling system in the SPES experimental facility

2014

The SPES (Selective Production of Exotic Species) experimental facility, under construction at the Italian National Institute of Nuclear Physics (INFN) Laboratories of Legnaro, Italy, is a second generation Isotope Separation On Line (ISOL) plant for advanced nuclear physic studies. The UCx target-ion source system works at temperature of about 2273 K, producing a high level of radiation (10^5 Sv/h), for this reason a careful risk analysis for the target chamber is among the major safety issues. In this paper, the obtained results of thermofluid-dynamics simulations of accidental transients in the SPES target cooling system are reported. The analysis, performed by using the RELAP5-3D 2.4.2 …

2018

The present article attempts to establish as accurately as possible the chronological trajectory of the unique codex of the Cancionero de Baena (PN1 in the Dutton nomenclature). It begins with a detailed examination of the codicological aspects of the manuscript, which serve to date its origin to around 1465. This origin, combined with the historical data, supports a conjecture that the manuscript probably belonged to Gonzalo de Beteta, an official of both Enrique IV and the Catholic Kings. It would have passed from him to his grandson, Jorge de Beteta y Cardenas, who gave it to the Real Biblioteca de El Escorial in 1576. The article then follows the vicissitudes of the manuscript from its …
