Descriptive study on subjective experience of genetic testing with respect to relationship, family planning and psychosocial wellbeing among women wi…

2021

AbstractBackgroundDue to increased risk of endometrial and ovarian cancer, women belonging to known Lynch Syndrome (LS) families are recommended to undergo germline testing. Current practice in Finland is to offer counselling to women with pathogenic variant and advocate risk-reducing surgery (RRS) after completion of childbirth. The present study aimed to clarify the impacts of positive germline testing on family planning and reproductive decisions of these women, which are relatively unknown.MethodsSeventy-nine carriers of germline MMR gene pathogenic variant (path_MMR)were identified from the Finnish LS Registry as having genetic testing performed before the age of 45 years and not havin…

Cognitive Performance and Psychological Distress in Breast Cancer Patients at Disease Onset

2019

ObjectiveMany cancer patients complain about cognitive dysfunction. While cognitive deficits have been attributed to the side effects of chemotherapy, there is evidence for impairment at disease onset, prior to cancer-directed therapy. Further debated issues concern the relationship between self-reported complaints and objective test performance and the role of psychological distress.MethodWe assessed performance on neuropsychological tests of attention and memory and obtained estimates of subjective distress and quality of life in 27 breast cancer patients and 20 healthy controls. Testing in patients took place shortly after the initial diagnosis, but prior to subsequent therapy.ResultsWhi…

CD40 activity on mesenchymal cells negatively regulates OX40L to maintain bone marrow immune homeostasis under stress conditions

2021

BackgroundWithin the bone marrow (BM), mature T cells are maintained under homeostatic conditions to facilitate proper hematopoietic development. This homeostasis depends upon a peculiar elevated frequency of regulatory T cells (Tregs) and immune regulatory activities from BM-mesenchymal stem cells (BM-MSCs). In response to BM transplantation (BMT), the conditioning regimen exposes the BM to a dramatic induction of inflammatory cytokines and causes an unbalanced T-effector (Teff) and Treg ratio. This imbalance negatively impacts hematopoiesis, particularly in regard to B-cell lymphopoiesis that requires an intact cross-talk between BM-MSCs and Tregs. The mechanisms underlying the ability of…

Stem Cell Therapy

2019

The necessity of improving the health and quality of life of subjects affected by diverse injuries and chronic diseases, has led to develop a new branch of translational medicine, called Regenerative Medicine (RegMed). RegMed has the principal objects of restoring, maintaining or enhancing structures and functions of tissues and organs. In order to achieve these goals, the RegMed researchers have proposed and tested diverse approaches. The Stem Cell Therapy represents the central focus of RegMed, even if its applications may imply the onset of adverse conditions. Here, an overview of this topic will be reported, by pointing advantages and disadvantages.

The diagnostic accuracy of PIK3CA mutations by circulating tumor DNA in breast cancer: an individual patient data meta-analysis

2022

Background: The circulating tumor DNA (ctDNA) diagnostic accuracy for detecting phosphatidylinositol 4,5-bisphosphate 3-kinase catalytic subunit alpha ( PIK3CA) mutations in breast cancer (BC) is under discussion. We aimed to compare plasma and tissue PIK3CA alterations, encompassing factors that could affect the results. Methods: Two reviewers selected studies from different databases until December 2020. We considered BC patients with matched tumor tissue and plasma ctDNA. We performed meta-regression and subgroup analyses to explore sources of heterogeneity concerning tumor burden, diagnostic technique, sample size, sampling time, biological subtype, and hotspot mutation. Pooled sensitiv…

Rak - osobliwe metafory choroby

2016

W artykule zostały przedstawione metafory, jakimi obecnie posługujemy się, by opisać chorobę nowotworową.

Liquid biopsy in colorectal carcinoma: the search for potential prognostic and predictive biomarkers

Background: Liquid biopsy is considered a repeatable, non-invasive and dynamic tool. It could be able to recover from liquid samples (expecially blood)) cancer-specific informations (miRNAs, circulating-free DNA (cfDNA), circulating tumor DNA (ctDNA) and exosomes) by overcoming the limitations associated with traditional tissue biopsy. Aim: to investigate the potential prognostic and predictive role of blood cancer-related biomarkers such as cfDNA or ctDNA and exosomal DNA in colorectal cancer (CRC) patients. Results: cfDNA or ctDNA and exosomal DNA could have a potential applicability in CRC management. New generation technologies are able to identify clinically relevant genomic alteration…

The Italian Society of Gastroenterology (SIGE) and the Italian Group for the study of Inflammatory Bowel Disease (IG-IBD) Clinical Practice Guideline…

2010

Biological therapies are an important step in the management of Inflammatory Bowel Diseases. In consideration of high cost and safety issues there is the need to have clear recommendations for their use. Despite the American Gastroenterological Association and the European Crohn's and Colitis Organisation have published exhaustive Inflammatory Bowel Disease guidelines, national guidelines may be necessary as cultural values, economical and legal issues may differ between countries. For these reasons the Italian Society of Gastroenterology and the Italian Group for the study of Inflammatory Bowel Disease have decided to elaborate the Italian guidelines on the use of biologics in Inflammatory…
