Metagenomics uncovers a new group of low GC and ultra-small marine Actinobacteria

We describe a deep-branching lineage of marine Actinobacteria with very low GC content (33%) and the smallest free living cells described yet (cell volume ca. 0.013 μm(3)), even smaller than the cosmopolitan marine photoheterotroph, 'Candidatus Pelagibacter ubique'. These microbes are highly related to 16S rRNA sequences retrieved by PCR from the Pacific and Atlantic oceans 20 years ago. Metagenomic fosmids allowed a virtual genome reconstruction that also indicated very small genomes below 1 Mb. A new kind of rhodopsin was detected indicating a photoheterotrophic lifestyle. The aim of this study was to determine the occurrence of tick-borne pathogens of medical importance in questing ticks collected from five recreationally used islands along the Norwegian coastline. Furthermore, since coinfection may affect the disease severity, this study aimed to determine the extent of coinfection in individual ticks or co-localization of tick-borne pathogens. In all, 4158 questing Ixodes ricinus ticks were analyzed. For detection of tick-borne encephalitis virus (TBEV), nymphs (3690) were analyzed in pools of ten. ‘Candidatus Phytoplasma solani’, belonging to the 16SrXII-A subgroup, is associated with grapevine Bois Noir (BN). It is usually transmitted by Hyalesthes obsoletus from a wide range of weeds to grapevine, which is considered a dead-end host. Moreover, other alternative vectors can play a role in the epidemiology of the disease. In Sicily, the presence of BN was reported in 1996, both on typical and on newly introduced cultivars. Furthermore, high BN incidence was observed on native grape cultivars grown close to ‘Chardonnay’ vineyards.
