Studies on the natural hemolytic system of the annelid worm Spirographis spallanzanii viviani (Polychaeta).

Abstract Hemolytic activity in the hemolymph of Spirographis spallanzanii was estabilished against Bufo, sheep, calf, rabbit, rat, human A, B, O erythrocytes. Chemico-physical treatments suggest that the hemolytic factor could be a thermo-labile protein whose activity requires calcium ions and which is active over broad pH and temperature ranges. Some of these properties and the sigmoidal curve obtained by plotting the quantity of hemolymph against the percentage of hemolysis, produce some analogies with the mammalian lytic system.

DNA fluorescence induced by polymethine cation pyrvinium binding

Pyrvinium is a polymethine cation which shows interesting fluorescence emission and DNA binding properties. In diluted aqueous solution, pyrvinium pamoate induced a bright yellow fluorescence in kinetoplast DNA from Trypanosoma cruzi epimastigotes as well as in chicken erythrocyte nuclei under a wide range of excitations. No fading was observed after mounting in suitable media. Spectroscopic studies on pyrvinium solutions revealed bathochromic and hypochromic shifts in the absorption spectrum of its complex with DNA. Subtypes of muscarinic receptor on cholinergic nerves and atrial cells of chicken and guinea-pig hearts

1. Electrically driven chicken and guinea-pig atria were used to investigate the negative inotropic effects of the muscarinic agonists methacholine and acetylcholine (ACh). The release of ACh from isolated hearts into the perfusate in response to (preganglionic) vagal or (pre- and postganglionic) field stimulation was bioassayed on the guinea-pig ileum or determined by labelling with [3H]-choline. 2. Concentration-response curves for the negative inotropic effect of methacholine were shifted to the right by pirenzepine in various concentrations (0.03 to 10 mumol l-1). Reproducing kernel hilbert spaces regression methods for genomic assisted prediction of quantitative traits.

Abstract Reproducing kernel Hilbert spaces regression procedures for prediction of total genetic value for quantitative traits, which make use of phenotypic and genomic data simultaneously, are discussed from a theoretical perspective. It is argued that a nonparametric treatment may be needed for capturing the multiple and complex interactions potentially arising in whole-genome models, i.e., those based on thousands of single-nucleotide polymorphism (SNP) markers. Mutation of the important Tyr-33 residue of chicken avidin: functional and structural consequences

The strong interaction between avidin and biotin is so tight (dissociation constant 10-15M) that conditions usually sufficient for protein denaturing fail to dislodge biotin from the avidin—biotin complex. This kind of irreversible binding hinders the use of avidin in applications such as affinity purification or protein immobilization. The effects of cold and glucagon on lipolysis, glycogenolysis and oxygen consumption in young chicks.

Abstract 1. 1. To study the possible role of glucagon in avian thermoregulation the effects of cold exposure and glucagon on lipolysis, glycogenolysis and oxygen consumption were measured in young chicks. 2. 2. Cold exposure (+10°C) and glucagon injection (0·3 mg/kg, i.p. at +30°C) both caused a marked increase in the plasma FFA and a decrease in the liver glycogen content. 3. 3. It is suggested that glucagon possibly acts in the avian thermoregulation by producing at least lipolysis and glycogenolysis during cold exposure.

Genome-wide analysis reveals the patterns of genetic diversity and population structure of 8 Italian local chicken breeds

The aim of this study was to conduct a genome-wide comparative analysis of 8 local Italian chicken breeds (Ermellinata di Rovigo, Millefiori di Lonigo [PML], Polverara Bianca, Polverara Nera, Padovana, Pepoi [PPP], Robusta Lionata, and Robusta Maculata), all under a conservation plan, to understand their genetic diversity and population structure. A total of 152 animals were analyzed using the Affymetrix Axiom 600 K Chicken Genotyping Array. The levels of genetic diversity were highest and lowest in PML and PPP, respectively. The ultrastructure of articular cartilage of the chicken's knee joint

The articular cartilage and synovial membrane of immature and mature chicken knee joints were studied by light, scanning and transmission microscopy. The findings differed from human articular cartilage and we conclude that the chicken knee joint is not suitable as a model for human joint degeneration.

The Process-inducing Activity of Transmembrane Agrin Requires Follistatin-like Domains

Clustering or overexpression of the transmembrane form of the extracellular matrix proteoglycan agrin in neurons results in the formation of numerous highly motile filopodia-like processes extending from axons and dendrites. Here we show that similar processes can be induced by overexpression of transmembrane-agrin in several non-neuronal cell lines. Mapping of the process-inducing activity in neurons and non-neuronal cells demonstrates that the cytoplasmic part of transmembrane agrin is dispensable and that the extracellular region is necessary for process formation. Determination of quinolone residues in chicken and fish by capillary electrophoresis-mass spectrometry

A specific pressure-assisted CE-MS method is described for the analysis of five quinolone residues. MS using a single quadrupole is compared with multiple-stage MS using a quadrupole IT (QIT-MS(n)). The procedure involves a common sample preparation by SPE on disposable cartridges. The most suitable electrolyte is 60 mM (NH(4))(2)CO(3) at pH 9.2. Single quadrupole does not provide enough fragmentation to confirm identities according to the current legislation. However, QIT-MS(n) achieves selective fragmentation.
